Objective:Matrix metalloproteinases is an important enzymes which involved in the extracellular matrix of synthesis and degradation.The MMP2and MMP9can degradate gelatin, Ⅳ and Ⅴ type collagen and elastic protein. They are in proliferative vitreoretinopathy forming process plays an important role. To observe the effect of Huyusanjie tablet on the Matrix metalloproteinnases2(MMP2) and Matrix metalloproteinnases9(MMP9) of retinal tissue of the experimental proliferative vitreoretinopathy animal model induced by Dispase IMethods:To inject0.1u/0.05ml Dipase I into intravitreous to induce the (PVR) animal model.the animals are divided into the blank control group, model control group, positive control group, Huyusanjie tablet high dose(HD) control groups, Huayusanjie tablet TD control groups,and Huyusanjie tablet low dose (LD) group.The Huyusanjie tablet control group were received different doses of Huyusanjie tablet of suspension liquid irrigation treatment stomach28days,the positive control group were received intravitreous injection of daunorubicin one-time.By the methed of Western Blot,to observe the MMP2and MMP9of retinal tissue.Results:(1) Develop the PVR animal model:blank control group compared with model control group in hyperplasia series, hyperplasia membrane cross-sectional area and section hyperplasia cell count, proliferating cell nuclear antigen (PCNA) is very significant differences (P<0.01).(2) After treatment7,14,21and28days, model control group compared with positive control group, Huyusanjie tablet HD control groups and TD control groups in hyperplasia series is very significant differences (P<0.01). After treatment7and14days, model control group compared with Huyusanjie tablet LD control groups in hyperplasia series is very significant differences (P<0.01).but is not differences (P>0.05)after treatment21and28days.(3) Model control group compared with treatment control groups in hyperplasia membrane cross-sectional area and section hyperplasia cell count is very significant differences (P<0.01).(4) Model control group compared with treatment control groups in proliferating cell nuclear antigen (PCNA) is very significant differences (P<0.01).(5)There is MMP2protein expression in every groups, The IOD value of MMP2of model control group is higher than the control group blank, there is a very significant difference (P<0.01); Model control group compared with positive control group,there is a significant difference (P<0.05), compared with Huyusanjie tablet HD control groups and TD control groups, there is a very significant difference, but compared with LD control groups there is not difference (P>0.05).(6) There is MMP9protein expression in every groups, The IOD value of MMP9of model control group is higher than the control group blank, there is a very significant difference (P<0.01); Model control group compared with Huyusanjie tablet HD control group.there is a very significant difference (P<0.05), compared with positive control group and Huyusanjie tablet TD control groups, there is a significant difference, but compared with LD control groups there is not difference (P>0.05).Conclusion:This experiment through the glass injection Dipase Ⅰ successful induced the PVR animal model. Huyusanjie tablet can restrain the cell proliferation and the expression Levels of PCNA,MMP2and MMP9of PVR rabit induced by Dipase Ⅰ.then play the preventive PVR development of function.
