Objective:To study the methods of suceessful creation of improved left varicocele model,To study the effect of improved left varicocele on the testis in rats,To evaluate its feasibility on the model.Methods:ELV models of SD rats were established by partial ligation of the left renal vein and ligation of the communicating branch of the left renal vein and the left common iliac vein in 21 SD rats, and another 15 were included in a sham operation group as controls.Three months after operation,rat′s spermatic vein′s diameter was measured by operating microscope. The testes were weighted by electronic scale.After producting testicular biopsy by HE, 10 seminiferous tubule break faces were observed in each sample.We assayed seminiferous tubule bore(D),peri-membrane depth(M),dissepiment germ cell′s layer number(P) and germ cell maturity(S).Every item′s full mark was 5,4 items accounted for20.The average score(TMS) also could be caleulated.Results:The left spermatic diameters in experimental group were significantly dilated than the right spermatic diameters and in control group(P<0.01).The left testieular weight in experimental group singificantly decreased compared with those of the right testieular weight and in control group(P<0.05). In the testis of experimental group,the seminiferous tubule bore(D) were diminuted, Peri-membranes (M) become thicker,cell layer number(P) decreased,germ cell's maturity(S) became distuthant. They had significant differences(P﹤0.01).Conclusion :The successful created experimental left varicocele rat model using improved method,which can be used for experimental study of the disease. Objective:To evaluate the effects of improved left varicocele(ELV) on testis enzymes and proliferating cell nuclear antigen(PCNA) in rats.Methods:ELV models of SD rats were established by partial ligation of the left renal vein and ligation of the communicating branch of the left renal vein and the left common iliac vein in 21 SD rats, and another 15 were included in a sham peration group as controls.At 3 months, the activity of the LDH,SDH and G6PDH were measured by olorimetry.The concentration of the PCNA was measured by enzyme-linked immunosorbent assay(ELISA).Results:The activity of the LDH and G6PDH (8.17±3.47,34.00±16.29)U/mg in left testes of experimental group were reduced remarkably than in right testes(12.69±3.97,78.03±25.28)U/mg and in control group(11.98±2.26,54.88±20.87)U/m(gP﹤0.05).The activity of the SDH was reducd remarkably in left testes of experimental group(1.22±0.41)U/mg than in control group(1.74±0.43)U/mg(P﹤0.05),but there were no significant difference between the left and right testes(1.62±0.56)U/mg in experimental group. The concentration of the PCNA was reduced in left testes of experimental group(669.10±205.39)mU/ml than in right teste(s800.81±172.02)mU/ml and in control group(776.81±231.72)mU/ml,but there were no significant differences(P﹥0.05).Conclusion :The reduction of the LDH,SDH and G6PDH and the changes of PCNA in improved ELV may play an important role in leading to subfertility.
