The Effect Of Defibrase On Fibronectin And Proliferating Ceil Nuclear Antigen In Traumatic Rabbit Eyes With Proliferative Vitreoretinopathy

OBJECTIVE:To study the effect of Defibrase on fibronectin and proliferating ceil nuclear antigen in experimental traumatic proliferative vitreoretinopathy.METHODS:Twenty rabbits were divided into experimental control and experimental groups at random. Anesthesia by intramuscular injection at rabbit stern. Then at the temple side 1:00 aspect, 3mm after the limbus of cornea, we injected toward the center of eye ball perpendicularly thought pupil under euthyphiria with 7# neddle, extracted 0.3ml vitreous body of the right eyes. 0.3ml blood extracted from the auricle vein was injected into the vitreous of experimental control group right eyes. 0.3ml blood extracted from the auricle vein and 0.1ml(0.06 unit) defibrase were injected into the vitreous of experimental group right eyes. The left eyes were not treated with anything as normal control group. Do not sut the cut, then droped 0.25% chloroptic into conjunctival sac, intramuscular injection penicillin(800 thousand unit) at rabbit stern. After 40 days ,observed retina by direct ophthalmoscope, then executed all the rabbits and enucleated the eyes immediately. Injected 10% formalin into vitreous body, then soaked into formalin liquid a week to fast the samples. A week later, cut open the eyes at the level of cornea and optic nerve, then alcohol dehydration, wax immerses, paraffin embedding, and then dyed the chip with HE and immunohistochemical SP routinely. The positive expression of FN,PCNA in the proliferative membranes were examined and the stage of PVR was evaluated..
