Objectives:To explore ways and means for promoting HPV infection of macrophagesMethods:1.Fresh genital warts were used for microparticles preparation and microparticleswere incubated with macrophages generated from peripheral blood mononuclear cells(PBMCs) of healthy donors. To collection of cultured macrophages and extract themicroparticles in the supernatant, such microparticles were further incubated withuninfected macrophages and then the macrophages were detected with HPV-DNA existenceby RT-PCR.2. Cellular immunochemistry was used for the detection of macrophagesco-cultured with microparticles are dyed brown.3. Electron microscope was used toobserve the existence of HPV within the nucleus of macrophages.4. Multiphoton-microscope was used for detection of microparticles within macrophages and HPVexistence within microparticles, as well as the ability of macrophages to uptake and releasemicroparticles.Results:1. RT-PCR showed that HPV6/11-DNA was positive in microparticle incubatedmacrophages.2. Cellular immunochemistry showed that HPV was positive withinmicroparticle incubated macrophages.3. Electron microscope revealed that there was HPVexistence within nucleus of macrophages that incubated with microparticles.4. Multiphoton-microscope imaging showed that after staining,the green macrophages containingred microparticles,containing green fluorescent staining HPV in the red microparticles, andlysosome was not colocalized with microparticles as well.Conclusions:1.HPV infection of macrophages by the way of microparticles2.HPVinfection of macrophages also produce and release a new microparticles containing HPV.
