Objective To investigate the expression of Wnt-1in patients with fibroid and cervical lesion (cervical intraepithelial neoplasia and cervical carcinoma).Methods161patients retrieved operation with pathologic diagnosis were collected randomly at gynecological department of the Shanxi Tumor Hospital from August2011to October2012.68cases of cervical of cervical squamous cell carcinoma,28cases of cervical intraepithelial neoplasia Ⅰ、Ⅱ,35cases of cervical intraepithelial neoplasiaⅢ(low lesion group28cases, HSIL group35cases), normal cervical tissue were randomly selected (over the same period due to the uterine fibroids rather than cervical lesions hysterectomy)30patients served as controls.Aged28to72years old, on average (36.50±1.592) years. All patients were without radiotherapy and chemotherapy after surgery confirmed by pathology. Staging of cervical squamous cell carcinoma FIGO2000clinical diagnostic criteria:68cases of cervical squamous cell carcinoma (which is divided into39cases of stage I clinical stage,20cases of a Phase Ⅱ,and9cases of a PhaseIII).We examined the expression of Wnt-1in patients by immunohistochemical SABC method,.at the same time,HPV is detect by PCR-reverse dot blot hybridization.Results1.HPV infection:HPV infection difference was statistically significant (χ2=38.443, P<0.001).The high degree of lesion group was higher than low-grade lesions group, the difference was statistically significant (P=0.013), cervical carcinoma is higher than in low-grade lesions group, the difference was statistically significant (P<0.001); the positive rate of cervical squamous cell carcinoma in highly than high degree of lesion group (P<0.001).2.Wnt-1immunohistochemical results:Wnt-1protein positive reaction is mainly localized in the cytoplasm, some can also be expressed on the cell membrane, positive cells were brown particles. Wnt-1protein strongly expressed in cervical squamous cell carcinoma. Wnt-1protein expression gradually increased.Each group of Wnt-1protein expression difference was statistically significant (P<0.001). The expression of Wnt-1protein of low lesions, high-grade lesions group, squamous cell carcinoma of the cervix were significantly higher than the normal group, the differences were statistically significant (P<0.001); HSIL group is highly than in the low-grade lesions group, the difference was statistically significant (P=0.048), cervical squamous cell carcinoma is higher than the low-grade lesions group, the difference was statistically significant (P<0.001); cervical squamous cell carcinoma group (P=0.039)were higher than that of high-grade lesions.3. There was significant positive correlation between the expression of Wnt-1and HPV infection (r=0.463, P<0.001),Conclusion1. Wnt-1protein in cervical carcinoma was significantly higher than that of cervical intraepithelial neoplasia (CIN) organization, at the same time the the HSIL expression in cervical intraepithelial neoplasia tissue was higher than low-grade lesions, low-grade lesions was higher than the expression of normal cervical tissue.2. HPV infection, the difference was statistically significant (χ2=38.443, P<0.001). The HSIL group was higher than the low-grade lesions group, the difference was statistically significant (P=0.013), and the positive rate of cervical cancer in low-grade lesions group, the difference was statistically significant (P <0.001); positive rate of cervical cancer in a high degree of lesion group (P<0.001).3. There is significant positive ceorrelation between the expression of Wnt-1and HPV infection。It indicates that Wnt signaling pathway may be actived by Wnt-1.
