Non-alcoholic fatty liver (NAFL) injury is the principal reasons of chronicliver disease. How to prevent and treat NAFL injury effectively has become one ofthe hotspots in the area of functional food and pharmaceutical research.Dendrobium huoshanense is a kind of valuable medicinal plant in China. Thepolysaccharides from D. huoshanense (PDH), which is one of the main activeingredients, possess the physiological active function according to the results ofmodern pharmacological researches. The effects of PDH on intervention andimprovement of NAFL injury had been studied in this paper.Firstly, the total PDH were obtained by the method of hot water and ethanolprecipitation. The contents of carbohydrates, total nitrogens, ash and themonosaccharide compositions in the total polysaccharides were analyzed. Secondly,the effects of the total PDH on intervention and improvement of NAFL injury in SDrats were obverved. For animal experiments, the SD rats were divided into fourteams including control team consisting of normal control group and high-fatcontrol group, intervention team consisting of normal with PDH intervention groupand high-fat with PDH intervention group, single improvement team consisting ofhigh-fat with PDH improvement group, high-fat with nicotinic acid improvementgroup and high-fat improvement group, and diet synergistic improvement teamconsisting of diet with PDH synergistic improvement group and diet with nicotinicacid synegristic improvement group. The indexes of organs, and biochemicalparameters in serum, lipids and antioxidation in liver of SD rats were detected, andthe changes in histological livers were observed at the end of the8thweek.The results showed that the contents of carbolydrates, total nitrogens and ashin the total PDH which composed of glucose, mannose and galactose in the molarproportions of31:10:8were84.12%,7.125%and1.191%, respectively. The resultsof animal experiments showed that compared with the normal control group, thecontents of TC, TG, ALT and AST in serum, the contents of lipids, TC, TG, HDL-C,LDL-C, MDA and H2O2in liver of high-fat control group increased significantly,but the content of GSH and the activities of SOD and CAT in liver decreased, which indicated that lipid peroxidation and vascular endothelial damage had happened tothe rats of high-fat control group. The levels of biochemical parameters in serum,lipids and antioxidation in liver of SD rats with high-fat diet and PHD werebetween that of the high-fat control group and that of the normal control group.There was no significant difference between the normal with PDH interventiongroup and the the the normal control group. The best improving effects wereobserved in the diet synergistic improvement groups. The results implied that thePDH could protect the SD rats from the anti-oxidative stress damage. The NAFLinjury could be effectively improved by the synergistic treatment of PHD and dietadjustment.In conclusion, the PDH as an adjunct to diet adjustment has the protective effectson NAFL injury, and the intervention and improvement mechanism may be relatedto balance of lipid metabolism restored by PDH.
