High-fat diet is one of the main factors that cause a variety of chronic diseases such as obesity,fat metabolism disorders,and "three highs".A large number of studies have found that a high-fat diet and irregular life can lead to the formation and development of non-alcoholic fatty liver disease(NAFLD).NAFLD has become the leading cause of chronic liver disease.Effective prevention and treatment of NAFLD is one of the research hotspots in the field of medicine and functional food.China began to study Inonotus obliquus in 1992.It is reported that Inonotus obliquus has biological functions such as blood sugar reduction,antitumor,antiviral,bacteriostatic,and immune regulation,and polysaccharide is one of the main components to exert its physiological function.In this thesis,a high-fat model of NAFLD mice was induced by a high-fat diet induction method.Inonotus obliquus polysaccharide(IOP)was used to induce basic growth indicators,blood lipid levels and antioxidant functions,liver function of high-fat diet NAFLD mice.The study of functional indicators and the effects of liver inflammatory response,explores the biological activity of Inonotus obliquus polysaccharide to protect the liver and its mechanism of action from the aspects of lipid regulation and inhibition of NAFLD liver injury.The main results are as follows:(1)Effect of Inonotus obliquus polysaccharide on basic growth indexes of NAFLD mice induced by high-fat dietThe results show that IOP can significantly reduce the body weight and liver coefficient of high-fat diet-induced NAFLD mice(P<0.01),and prevent large amounts of fat from accumulating in mice.Compared with the high-fat model group,the high-dose(200mg/kg)polysaccharide from Inonotus obliquus had the best effect,reducing mouse body weight by 12%,liver coefficient by 21.3%,and fat coefficient by 35.8%.The results suggest that IOP can significantly improve the body weight and liver fat accumulation of NAFLD mice.(2)Effects of Inonotus obliquus polysaccharide on blood lipid levels and antioxidant function in NAFLD miceStudies have found that Inonotus obliquus polysaccharide can significantly improve the hyperlipidemia and reduced antioxidant capacity of NAFLD mice induced by a high-fat diet(P<0.01).Compared with the high-fat model group,the high-dose(200mg/kg)polysaccharide from Inonotus obliquus reduced the mice TC level by 29.2%,TG by 57.0%,LDL-C by 48.3%,and HDL-C by 18.9%,the atherosclerosis index AI decreased by 82.2%,the content of T-SOD,T-AOC,GSH-Px,and CAT increased by 31.4%,57.1%,38.5%,and 29.1%,and the MDA content decreased by 25.5%.The results suggest that Inonotus obliquus polysaccharide has the ability to prevent atherosclerosis in NAFLD mice and enhance their ability to resist lipid peroxidation.(3)Effect of Inonotus obliquus polysaccharide on liver function indexes of NAFLD miceStudies have found that Inonotus obliquus polysaccharide can significantly reduce the activity of serum transaminase,reduce the content of serum bilirubin,and increase the content of hemoglobin(P<0.01).Compared with the high-fat model group,high-dose(200mg/kg)intragastric administration of IOP decreased the ALT,AST,and ALP activities in NAFLD mice by 40.2%,40.3%,and 31.3%,TBIL decreased by 63.1%,and DBIL decreased by 47.6%,total protein increased by 48.8%,and albumin increased by 35.0%.The results showed that the Inonotus obliquus polysaccharide can effectively improve the abnormal liver function indexes and reduce the degree of liver damage in NAFLD mice caused by high-fat diet.(4)Effect of Inonotus obliquus polysaccharide on liver inflammation in NAFLD miceThe results show that high-dose(200mg/kg)Inonotus obliquus polysaccharide can significantly reduce the content of liver inflammatory factors.Compared with the high-fat model group,the levels of TNF-?,IL-6,and IFN-? decreased by 27.9%,47.7%,and 11.3%(P<0.01).The results indicated that Inonotus obliquus polysaccharide can reduce liver inflammation and repair liver damage by adjusting the content of liver inflammatory factors in NAFLD mice.In summary,Inonotus obliquus polysaccharide can significantly reduce the body weight and liver coefficient of NAFLD mice induced by a high-fat diet,improve the symptoms of hyperlipidemia and weakened antioxidant capacity,reduce the activity of serum transaminase and the content of serum bilirubin,increase hemoglobin content,reduce liver inflammatory factors TNF-?,IL-6 and IFN-? content.Inonotus obliquus polysaccharide have biological activities of reducing blood lipids,preventing atherosclerosis,and protecting the liver.
