Chicory belongs to Asteraceae.Its root is fleshy and short.It is a medicinal and edible plant.Chicory is mainly distributed in Europe,Asia and North Africa.Chicory is rich in bioactive substances such as polysaccharides,phenolic acids,alkaloids,terpenes and so on.Chicory polysaccharide is a crude polysaccharide extracted from chicory root and separated from the root of chicory.Two new polysaccharides are purified by cellulose column and glucan column,which are named CP-1 and CP-2.Some studies have shown that chicory polysaccharides have antioxidant,anti-inflammatory,hypoglycemic and hypolipidemic functions.However,the effect of purified CP on nonalcoholic fatty liver disease in male rats has not been reported.In this study,the antioxidant activity of CP-1 isolated from CP,anti fatigue and lipid lowering effect on NAFLD rats were studied to provide scientific theoretical basis for the rational exploitation and utilization of chicory polysaccharides.High fat diet rats were given different doses of CP-1 for 8 weeks after modeling for 4 weeks.The levels of HLD,LDL,VLDL,GLU,ALP,SOD,MDA,TG,TC,ALT and AST in serum and liver homogenate were determined by taking serum and a certain amount of liver homogenate.The expression level of inflammatory factors,lipid metabolism related transcription factors and downstream genes,the total amount and phosphorylation level of AMPK alpha and downstream molecules of lipid metabolism signaling pathway,HE staining liver tissue sections were observed,and ultrastructure of liver cells was observed by transmission electron microscopy.The results showed that compared with model rats,oral CP-1 significantly decreased body weight and liver index.CP-1 can also significantly increase the serum SOD and HDL-C levels of NAFLD rats,and reduce the contents of ALT,AST,TG,TC,LDL-C,GLu,ALP,and LDL-C.At the same time,CP-1 can also effectively reduce the levels of MDA,TC and TG in the liver of NAFLD rats,and increase the levels of SOD and T-AOC in the liver.In addition,CP-1 can enhance the expression of ATGL,CPT-1 and P-ACC by reducing the expression of AMPK,and reduce the expression of FAS,ACC and SCD-1 in liver.Therefore,CP-1 can alleviate NAFLD induced by high-fat diet.The results of antioxidant activity in vitro showed that when the concentration of CP-1 was 2.4 mg/mL,the hydroxyl radical,oxygen free radical and DPPH radical scavenging rate were 40.93%,67.11%and 58.17%respectively,while the reduction ability was slightly lower than that of the same concentration VC.The results of anti fatigue study showed that compared with the blank group,the weight swimming time of the oral CP-1 group was obviously prolonged,and the content of blood urea nitrogen(blood urea nitrogen,BUN),blood lactic acid(blood serum lactic acid,BSLA)and malondialdehyde(malonaldehyde,MDA)decreased obviously.The levels of hepatic glycogen(HG)and muscle glycogen(MG)increased significantly.The results showed that CP-1 had strong antioxidant and anti fatigue effects.
