Nonalcoholic fatty liver disease is one cause of fatty liver,occurring when fat is deposited(steatosis)in the liver due to endogenous or exogenous causes.Currently NAFLD has become the second to the viral hepatitis liver disease,with complex causes,the onset of infection in people widely spread and easily lead to cirrhosis.Along with economic development and the influence of lifestyle,nonalcoholic fatty liver disease has become one of the most important public health problem in today's world.Grifola frondosa is a kind of precious edible and medical fungi.The G.frondosa polysaccharides have the functions of antioxidant,anti-aging,antitumour,enhance immunity,antivirus,antibacterial,etc.The chemical constituents of maitake included multivitamin,mineral substance and bioactivator.The G.frondosa polysaccharides play an important role in these function.In this study,the G.frondosa were used as material.First,we optimized the extraction technology of polysaccharides.Second,we investigated the protective effect of the polysaccharides from Grifola frondosa fruits on non-alcoholic fatty liver disease,then we compare them with chitosan.The experimental results are as follows:First,in this study,we optimized the extraction method,inserted complex enzymatic in maitake with ultrasonic method.The optimum extraction experiments focus on ultrasonic power,reaction time,reaction pH value,reaction temperature.By single factor experiment and orthogonal experiment,we obtained the best condition at 300W ultrasonic power,enzymatic time at 60min,enzymatic pH value of 7.5,reaction temperature at 40?.Polysaccharide extraction rate was 7.39%.Second,this experiment was performed by feeding high-fat feed mice caused by chronic mouse model of non-alcoholic fatty liver disease,by giving them Grifola frondosa polysaccharide and chitosan lavage,compared with model group,maitake polysaccharide and chitosan drug groups showed mitigated lesions.High dose groupGrifola frondosa polysaccharides had a significant effect(P<0.05)in restoring liver cell activity.Serum biochemical tests analysis and Grifola frondosa polysaccharides and the effect of chitosan on fatty liver in mice were compared,concluding that Grifola frondosa polysaccharide with chitosan can significantly influence the mice serum ALT,AST,TC,TG,HDL-C,LDL-C(P<0.05),inhibit the accumulation of fat in the liver of mice.Grifola frondosa polysaccharide high dose group in reduced serum ALT,AST and TG has a better effect even than the same doses of chitosan.However,chitosan effect on TC and HDL-C is better than Grifola frondosa high dose group at same levels.The above study have shown that Grifola frondosa polysaccharide can significantly improve liver function,especially maitake polysaccharide with high dose group obviously improved the liver function of non-alcoholic fatty liver disease,reduced triglyceride and cholesterol in the blood,but their ability of processing lipid and cholesterol is lower than chitosan.Perhaps this results are due to maitake polysaccharide isolation or the individual differences of the mice,although the preliminary experiment proved that maitake for prevention of non-alcoholic fatty liver disease has a certain effect and the effect of maitake high-dose group(500mg/kg)is similar to chitosan high-dose group.According to our results we concluded that Grifola frondosa polysaccharides have a good effect protecting mice liver besides improves quality of life.
