Objective:To observe the Lycium barbarum polysaccharides on the alcoholic fatty liver in rats of the preventive effect and to explore its mechanism.Methods: 65 male Wistar rats of clean grade were randomly divided into 4 groups: normal control group (15), alcohol group (20), the prevention of Lycium barbarum polysaccharides of low-dose group (15), the prevention of high-dose barbarum polysaccharide group (15). Normal control group gavaged with distilled water a total of 10 weeks. Alcohol gavaged alcohol group: 5 weeks ago to give the volume fraction of 40% (V / V) (relative density of ethanol 0.949 g / ml) of ethanol 8g/Kg.d (that is, 21.1 ml / kg.d) , from the sixth week to give the volume fraction of 50% (V / V) (relative density of ethanol 0.931 g / ml) 9g/Kg.d (that is, 19.3ml/kg.d) oral ,to the tenth week..Drug prevention group gavaged in ethanol (method ibid.) increase of the drug administered at the same time: the prevention of low-dose group Lycium barbarum polysaccharides Lycium barbarum polysaccharides (LBP) 250mg/kg/d (ie 5% of LBP solution - using saline configuration ); the prevention of high-dose group Lycium barbarum polysaccharides Lycium barbarum polysaccharides (LBP) 500mg/kg/d (that is, 10% of the LBP solution - configuration with normal saline); oral dose of 0.5 ml/l00 g, a total of 10 weeks . each group were killed over the weekend, compared 4 groups of rats: By oil red 0 staining of liver pathology light microscope morphology, determination of their liver cells and what the size of lipid droplets. Routine determination of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST),γa acyltransferase glutamyl (γ-GT), using enzyme-linked immune sorbent assay (ELISA method) for determination of serum leptin level.Results: the control group show the structure of the hepaic lobule is clear, a very small number of liver cell lipid droplets, and red is located in the lipid droplets in liver cells, alcohol group of red lipid droplets markedly increased. Drug prevention group the two red lipid droplets varying degrees of reduction. The form of lipid droplets mainly bullous type. Light microscopy, lipid droplets mainly bullous see the rats only a few: the control group 0, the alcohol group of 12, the prevention of low-dose group 4, high-dose prevention group 4; light microscopy, lipid droplets larger than 30% of the only the number of rats: control group 0, 13 alcohol group, the prevention of low-dose group 4, the prevention of high-dose group 4. The prevention of alcohol and the two groups more than the normal group, there was a significant difference (P <0.05); the two prevention groups less than the alcohol group, there are significant differences (P <0.05); the two groups compared to the prevention of non-significant difference (P> 0.05). The prevention of alcohol and two levels of serum ALT content than the control group showed no significant difference (P> 0.05); the prevention of alcohol and two levels of serum AST,γ-GT were significantly increased, and there are significant differences in the control group (P <0.05 ); the two groups than those in the prevention of alcohol serum AST,γwere significantly lower a GT, there are significant differences (P <0.05); prevention group compared to the two non-significant difference (P> 0.05). The prevention of alcohol and two levels of serum leptin concentration was significantly increased, and there are significant differences in the control group (P <0.05); the two groups than those in the prevention of alcohol group were significantly lower serum leptin, a significant difference (P <0.05); two prevention group compared to no significant difference (P> 0.05).Conclusion : Lycium barbarum polysaccharides are effective prevention and treatment of alcoholic fatty liver in rats may be related to by reducing the level of leptin.
