Objective: to study the effect of Xuelian injection on the different subtype of NOS aftercerebral ischemia/reperfusion in mice and investigate its antioxidant mechanism.Method:72male Kunming mice are divided into four groups, sham group(n=18), vehiclegroup(n=18), Xuelian injection group(n=18) and edaravone group(n=18). Each group ofmice were given Xuelian injection (0.6ml/kg/d), edaravone injection (3mg/kg/d) and equalvolume of plasmin (saline) by intraperitoneal injection for seven days, and then the middlecerebral artery occlusion model was established. The Neurological deficit score wasmeasured after MCAO for24hours.The volume was evaluated by TTC after MCAO for24hours. The different subtype of NOS was detected by immunohistochemistry andRT-PCR after MCAO for24hours. Result:(1)As same as edaravone injection, Xuelianinjection could improve Neurological deficit after MCAO for24hours(P<0.05,compared with saline group).(2)Xuelian injection also could decrease infarct volume afterMCAO for24hours(P<0.05, compared with saline group).(3)The same as edaravoneinjection, Xuelian injection could reduce the positive cells of iNOS and nNOS in infarctside, down-regulate the expression of iNOS and nNOS mRNA, increase the positive cellsof eNOS in infarct side after MCAO for24hours (P<0.05, compared with saline group).Conclusion: As same as edaravone injection, Xuelian injection could play play aneuroprotection after cerebral ischemia/reperfusion, through down-regulated theexpression of iNOS and nNOS, increased expression of eNOS after the cerebralischemia/reperfusion.
