Objective:To study the analgesic effect of plantar incision painand after intraperitoneal injection?ip.?of Xuelian injection in rats and the expression of SP?NK-1R in spinal marrow of rats.Methods:96 male adult SD rats were randomly allocated to 3 groups?n=32 each?:tramadol injection group?group T?,Xuelian injection group?group S?and control group?group C?,rats received operation to establish the model of plantar incision pain.Rats in groups T?S and C respectively received intraperitoneal injection of Tramadol 10 ml/kg,Xuelian injection 0.72 ml/kg,equal volume of normal saline.Paw withdrawal mechanical threshold?PWMT?was recorded at 1 h?T1?,2 h?T2?,4h?T3?,8 h?T4?after dosing,and then rats were sacrificed,then blood was collected and spinal cords?L4-6?were removed for detection of the changes of substance P?SP?and neurkinin-1 receptor?NK-1R?in serum by ELISA,and protein expressions of SP and NK-1R in spinal cord of rats by Western blot.Results:There was no difference in PWMT between the groups before surgery.PWMT decreased in each group than that before surgey?P<0.05?,there was no statistical difference between group S and groupT?P>0.05?.Compared with group C,PWMT were significantly increased in group S and group T?P<0.05?.Compared with group C,Level of SP and NK-1R in serum and protein expression in spinal cord of group S and group T were decreased and down-regulated?P<0.05?.There was no significant difference between group S and group T?P>0.05?.Conclusion:Xuelian injection and tramadol injection showed equal analgesic effect in incision pain rats.It is feasible for Xuelian injection to replace tramadol as analgesia choice for moderate and severe pain after surgery.
