| Objective:By investigating the expression of E-cadherin in bleomycin-induced oral submucos fibrosis (OSF) model before and after hyperbaric oxygen therapy (HBO) to explore the effect of HBO on bleomycin-induced OSF model.Methods:36Sprague-Dauley (SD) rats were randomly divided into7groups (2cases in experimental group1,6cases in experimental group2, experimental group3group each,7cases in experimental group4,5cases in control group1,control group2,normal group each). Rats in the experimental group and control group were injected BLM diluent to the bilateral buccal mucous membrane under lsoflurane (inhalation) anesthesia, once a day for6weeks. Rats in the experimental group were subjected to HBO(2ATA,60min/d,10d) after the model was made. Rats in experimental group1were executed after HBO had been done, experimental group2, experimental group3, experimental group4, control group2,nomal group were executed15days,30days,45days,45days,45days respectively after experimental groups taken HBO. Rats in control group1, were executed after the model was made. HE staining were used for detecting changes of the histopathological features and SABC immunohistochemistry was used for detecting the expression of E cadherin.Results:1. Local injection of1mg/ml BLM diluent to the bilateral buccal mucous membrane of SD rats, once a day for6weeks, can create similar changes with human OSF in the way of histopatholagical changes, without significant pathological change in45days after HBO;there were2cases with atypical hyperplasia.2. A positive expression of E-cadherin was detected in normal oral mucosa; significant decreasing of expression of E-cadherin could be found in BLM-induced OSF model(P<0.05).3. There was no significant difference between experimental groups and control groups in expression of E-cadherinConclusions:1.Local injection of1mg/ml BLM diluent to the bilateral buccal mucous membrane of SD rats, once a day for6weeks, can create similar changes with human OSF. Model made in this way was of stability and repeatability; Atypical hyperplasia may exist in this model.2. Compared with the nomal mucous membrane, there were significant decreasing of expression of E cadherin,which indicated that decreasing of expression of E cadherin may contribute to the pathogenisis of BLM-induced OSF model. 3. There were no difference in expression of E-cadherin after HBO (2ATA,80minQd,10d),which indicated that EMT,45days after HBO, was of little anti-fibrosis effect on BLM-induced OSF model. |
