Objective:this research through the test high-risk type Human Papillomavirus (high-risk types of Human Papillomavirus, HR-HPV) load and PCNA (Proliferating Cell Nuclear Antigen, PCNA) expression in different cervical lesions, to discusse the correlation among those.Methods:the second generation of hybrid capture technology (HC Ⅱ) is used to test the road of HR-HPV in23cases of chronic cervicitis,41cases CINI,37cases of CINⅡ,43patients with CINⅢ,34cases of cervical cancer patients; Application immunohistochemical techniques in the protein level detection of PCNA expression. Analyzing the correlation between both of them and cervical lesions.The results:1)HR-HPV in chronic cervicitis, CINⅠ, CINⅡ, CINⅢ, cervical cancer were the were13.0%,34.1%,81.1%,88.2%,88.4%, with the positive rate in the CINII, CINIII, cervical cancer is higher than this in chronic cervicitis (P<0.01), the positive rate among CINII, CINIII, cervical cancer is no difference (P>0.05).2) proportion of patients with chronic cervicitis HR-HPV load in0-<1pg/ml,1~<10pg/ml,10~<100pg/ml,>100pg/ml interval period is respectively87.0%,4.3%,8.7%and0.0%; in patients with CINI respectively65.9%,7.3%,17.1%and9.8%; in patients with CINII respectively18.9%,5.4%,29.7%and45.9%; in patients with CINIII respectively11.6%,4.7%,9.3%and74.4%; in patients with Cervical cancer respectively11.8%,5.9%,14.7%,67.6%. HR-HPV load in CINII, CINIII and cervical cancer group is significantly higher than chronic cervicitis group and CINI group, with a statistical significance (P<0.001). HR-HPV load in CINII is higher than CINI group, with a statistical significance (P>0.05). There is no difference between cervical cancer and CINIII (P<0.05).3) HR-HPV load and cervical lesions was positively related degree, R=0.607, P=0.000, with higher degree of cervical lesions, HR-HPV load of the increased gradually.4) proportion of patients with CINII or above in HR-HPV>100pg/ml is higher than those1~<10pg/ml,10~<100pg/ml(P<0.005), and this is no difference between the period of1~<10pg/ml and10~<100pg/ml (P>0.05). Probability of CINII or above in the patients whose HR-HPV load frequency>100pg/ml is higher.5) PCNA in chronic cervicitis, CINI, CINII, CINIII, cervical cancer patients were8.7%,12.20%,59.5%,84.7%,94.10%. The expression of PCNA enhanced with cervical lesions increasing. Expression of PCNA in CINII, CINIII and cervical cancer group is higher than those in chronic cervicitis group and CINⅠ, with a statistical significance (P<0.001).6) HR-HPV load and the expression of PCNA are positively related, R=0.678, P=0.000, with HR-HPV load increase gradually, the expression of PCNA gradually strengthened.7) single HR-HPV sensitivity test is72.3%, and the specific degrees for87.0%, up to about0.593index; PCNA sensitivity test for61.3%alone, the specific degrees for91.3%, up to about0.526index; Both united detection sensitivity was78.1%, the specific degrees for87.0%, up to about0.651index.Conclusion:1With cervical lesions increased degree, the amount of HR-HPV load increases gradually. The incidence of CINII significantly higher with the HR-HPV load≥100pg/ml2with cervical lesions increased degree, PCNA and HR-HPV load increase.
