Background:HPV infection is the main cause of cervical cancer. Cervical intraepithelialneoplasia is a precancerous cervical lesions, it develops by abnormal cells’ continuousabnormal proliferation and finally breaking through the basement membrane lead to theoccurrence of cervical cancer. With the content of high-risk HPV load and cervicallesions, two cell adhesion molecules analysed,we study the machaism of the progess ofcervical lesions, and try to provide the basis for the ultimate elimination of cervicalcancer.Objective:Study the mechanism of CIN progress by investigating the link among High-riskHPV load and cervical lesion level, E-cadherin protein and Numb protein expression.Process:Choose63cervical tissue specimen of paraffin wax cases of patients with chroniccervicitis,CIN or cervical cancer (chronic cervicitis in13, CINI-II in11cases, CINIII in16case,cerical cancer in23).Detect the E-cadherin protein,and Numb proteinexpression by the immuno-histochemical method. Test every patient’s HPV load withthe second generation postoperatively in hybrid capture (hybrid capture II, HC II).Result:1In63cases of cervical lesions specimens, with the increase of cervicallesions degree, hr-HPV load median value also increased. The chi-squaretest results of hr-HPV virus load group shows that it has obvious significantwith cervical degree levels(p=0.015<0.05), Spearmans rank correlationanalysis make it clear that they are positive correlated at the level ofcoefficient r=0.472. 2In the63cases’ specimens, the high expression rate of E-cadherinprotein in inflammation group,CINI-II, CINIII and squamous cell carcinomagroup of respectively were76.92%(10/13)54.55%(6/11),37.50%(6/16),17.39%(4/23), show that with the increase of cervical lesions degree,highexpression rate drop. Application a chi-square test results show that theE-cadherin protein in different levels express differently with statisticalsignificance(p=0.014<0.05).3E-cadherin protein in inflammation group, CINI-II, CINIII andSquamous cell carcinomain group of Cell membrane expression rate are:75%(9/12),70%(7/10),31%(4/13),15%(2/13), cytoplasm expression rateare:25%(3/12),30%(3/10),69%(9/13),85%(11/13), show that with theincrease of the lesions degree, the Cell membrane expression graduallyreduce, instead of the cytoplasm expression gradually increasingsignificantlywith the application of the chi-square test.(p=0.001<0.05).4The high expression rate of Numb protein in inflammation group,CINI-II, CINIII and squamous cell carcinoma group are7.69%(1/13),9.09%(1/11),51.25%(5/16),47.83%(11/23), show that with the increase ofcervical lesions level, high expression rate gradually increasing,Applicationa chi-square test, the statistical is significance.(p=0.039<0.05).5The high expression rate of E-cadherin protein in different viral loadgroup less than100RLU/CO,100-1000RLU/CO, more than1000RLU/COrespectively were55%(12/22),44%(11/25),19%(3/16), Spearman rankcorrelation analysis showed the level coefficient r=0.365,The highexpression rate of Numb protein in different viral load group respectivelywere19%(4/22),44%(6/25),50%(8/16), Spearman rank correlationanalysis, level coefficient r=0.345,There were positive correlation betweenthe virus load and both of the two protein.Conclusion:1. With the progess of cervical lesions degree, hr-HPV virus loadmedian value is increases,with hr-HPV virus load after group,show that thehigher the infection virus load,the higher possible suffering from high levelof cervical lesion,but there are no critical values be found for different CINdegree,so the histological examination must be combined.2. With the progess of cervical lesions, the expression of E-cadherin protein falls in cell membrane and increase incytoplasm untill extending toall of the epithelium,this shows the adhesion with cells abate gradually.Numb protein expresses mainly in cytoplasm, and increases with theprogess of cervical lesions.3. There are significant correlation between the the hr-HPV load with E-cadherin protein and Numb protein.Because of the HPV infection,the cellsmicro-environment changes along with the HPV virus load increasing, theadhesion between the cell abates which promoting the progess of the disease,but there is no significant related coefficient, This shows that besides theHPV infection, there are probably other causes and mechanism involved inregulating cell micro-environment changed.
