Background and Objective:Rectal cancer is a common malignant tumor of the alimentary canal.In recent years,neoadjuvant therapy and gene therapy, immune therapy, targeted therapy to rectal cancer is on extensive research.Along with to the tumor cell signal transduction pathways the deepening of the research, the molecular target therapy for the cell receptors, signal transduction, the cell cycle and angiogenesis, has become a new direction of cancer treatment. The ASC gene is a concerned genes in recent years which associated with cell apoptosis. Researches have proved existing ASC gene promoter of frequent methylation in breast cancer, prostate cancer, lung cancer, gastric cancer and other tumors, resulting in reduced expression of ASC gene,and thus lead to cancer through a variety of pathways. But the research of expression of ASC in rectal cancer is still lacking.This research detect the protein expression of ASC in carcinoma of rectum and other related paraneoplastic tissues,explore its correlation with clinicopathological characteristics.Methods:Forty cases rectal carcinoma resection specimen of the Second Affiliated Hospital of Shandong University of Traditional Chinese Medicine from2006to2009. All patients were not treated with chemotherapy and radiation before surgeon. To detect the ASC in colorectal cancer and normal tissue adjacent organization of positive expression rate by immunohistochemical techniques conventional S-P method, annalyze the relation of the expression of ASC and rectal cacinoma TNM stage,histological differentiation.We had negative control by repeating the same procedure with PBS balanced solution replacing No.l antibody,and had positive control by repeating the same procedure with positive slice containing objective antigen.According to the clinical data confirm its sexuality, age, differentiation and the clinical stages, and separately as grouping variables, protein level expression as index variables, make the statistical analysis.Results:The positive expression of ASC in rectal carcinoma tissue and adjacent normal tissues were respectively17.5%(7/40) and65.0%(26/40), P<0.01, there are significant differences. The positive expression of ASC in the TNM stage I and II was17.6%(3/17) and it in stage III, IV was17.4%(4/23), they were no significant differences. The positive expression in low differentiated carcinoma was4.8%(1/21), obviously lower than it in well-differentiated interferon-gamma31.6%(6/19), P=0.026.Conclusions:The positive expression of ASC in rectal carcinoma tissue were significantly lower than it in the adjacent normal tissue, their differences were significant. The expression of ASC in rectal carcinoma tissue was not related with patients gender, age, tumor stage,but histological differentiation degree are concerned. It clew ASC may affect the rectal cancer incidence, apoptosis and developing process.
