Objective:The rectal carcinoma is one of the most common tumor types in our country.In present,scientists focus their studies on cancer surppressor genes to find out the moleculor mechanisms of tumor oncogenesis. The purpose of this study is to observe The significance of the expression of NDRG2 and NDRG4 gene in carcinoma of the rectum and correlative research.Look forward to exploring the target therapy of molecular level from the rectal cancer surgery from molecular level.Method:Select the First Affiliated Hospital of Anhui Medical University in 2009-2010 years between surgery and pathologically confirmed in 44 cases of low rectal cancer specimens as the experimental group,all patients were not receiving chemotherapy,radiotherapy or other anti-cancer therapybefore surgery,and a complete follow-up data.The 44 patients taking the corresponding adjacent normal tissues as control group.Were measured by the immunohistochemical SP method staining tumor tissue and adjacent normal tissue NDRG2 and NDRG4 expression levels.Analysis of these indicators and rectal cancer size, depth of infiltratio,histological type,lymph node metastasis and Dukes staging. Statistical analysis:All the data were analyzed with SPSS13.0 statistics software.The statistical significance was defined as P<0.05.Result:1,In 44 cases of low rectal cancer tissues, NDRG2 and NDRG4 expression level was significantly lower than in normal adjacent tissues,there was a significant difference between two groups(P<0.05).Positive expression rates of NDRG2 and NDRG4 were found 54.5%(24/44) and 59.1%(26/44) in rectal carcinoma tissues,significantly lower than 93.2%(41/44) and 90.9%(40/44) in normal adjacent tissues evidently.2,In all positive expression cases,there were 22 cases of NDRG4 in 24 cases of NDRG2, both positive expression rates were 50%(22/44),there were close compatibility among NDRG2 and NDRG4 in rectal carcinoma(r=0.587).3,The expression of NDRG2 and NDRG4 in rectal carcinoma was closely related to the degree of differentiation and lymph node metastasis, there was a significant difference between two groups(P<0.05).The positive rate of NDRG2 and NDRG4 in the well- and moderate- differentiation group was higher than in the poor- differentiation group,and in no lymph node metastasis group was higher than in lymph node metastasis.4,The expression of NDRG2 and NDRG4 in rectal carcinoma was not correlated with age,sex, tumor size,depth of infiltratio,as well as the Dukes stage (P>0.05).Conclusion :1,NDRG2 and NDRG4 play an important role in the development in the incidence of rectal carcinoma,both of abnormal expression influence the level of the normal protein,s expression,lead to the incidence of rectal cancer direct or indirect.We suggest NDRG2 and NDRG4 may be plays a inhibitive role by their signal channels of rectal cancer,s formation.2,Compatibility among NRDG2 and NDRG4,suggesting that the two may be synergies in the occurrence and development process of rectal carcinoma.3,The expression of NDRG2 and NDRG4 are associated with rectal tumor's pathologic biologic character. The lower Tumor degree and lymph node metastasis,the less expression of NDRG2 and NDRG4.This prompted the two may suppress the occurrence and the development process of rectal carcinoma.4,The expression of NDRG2 and NDRG4 in rectal carcinoma was not correlated with age,sex, tumor size,depth of infiltratio,as well as the Dukes stage, suggesting that there may be independented in the occurrence and development process of rectal carcinoma.5,NDRG2 and NDRG4 high expression in some way is the suppression of lymph node micrometastases in colorectal cancer cases, suggesting NDRG2 and NDRG4 likely targeted therapy can be used as the molecular basis of cancer.
