Objective:The proliferation and differentiation of adipocytes can cause an increase in the number of fat cells, becoming larger, the excessive accumulation of fat particles. they result energy metabolism imbalance causing obesity and related diseases,a huge economic burden to the social and economic development. Environmental factors is one of the factors for obesity disease. Phthalate esters (PAEs) are persistent organic pollutants,they have environmental estrogen-like effect, and there is a higher exposure in the human body.But now studies are concentrated in the reproductive toxicity hazard on these toxicity. Report in the literature and preliminary research suggest that obesity-related PAEs have associate with abdominal obesity and children obesity, but no impact mechanism.Methods:This Experiment subject PAEs peroxisome proliferator agent as the starting point, PAEs can activate the peroxisome proliferator-receptors(PPARs). Studying DBP and DEHP which are China’s main industrial used and high exposured in body.In vitro culture establishment of experimental models of3T3-L1preadipocytes for mouse, the classic hormone cocktail induced differentiation of3T3-L1adipocytes, design two kinds of logo the PAEs exposure to participate in3T3-L1preadipocytes induced to differentiate, morphological changes were observed under an inverted fluorescent microscope with oil red O staining,lipid droplet formation process and the number of lipid droplets change; oil red O extraction of quantitative analysis of changes in fat cell lipid droplet content; real-time fluorescence quantitative PCR analysis of differentiation gene PPARy and adiponectin expression changes.Results:In the second day of the3T3-L1induced differentiation, the exposed dose group appeared lipid droplets firstly. when induction of lipid droplets of differentiation ended, the exposure dose group content greater than the blank control group. At the same time, different exposed dose group of lipid droplets increased with increasing exposure dose. Adipocyte differentiation genes of PPARy and adiponectin increased with the increase of exposure dose(P<0.01).Conclusion:PAEs is a synthetic peroxisome proliferator (peroxisome proliferators, PP). It can combine with adipocyte differentiation genes PPARy to be activated. PPARy involved in adipocyte differentiation process. Both Adipocyte differentiation genes of PPARy and adiponectin were up regulated, so that mature fat cells and theses fat particles increased significantly. the results provides a new way to study relation the PAEs of obesity, a scientific basis for public health policy formulation. And open new avenues for future obesity prevention and treatment of diseases, obesity and environmental pollution. |
