| Objective:To measure the concentration of four kinds of phthalates esters metabolites in urine of the pregnant women in TianJin area by using LC/MS method, and evaluation the exposure level of pregnant women; To analyze the influence on the concentration of PAEs by lifestyle; To detect the expression of three steroidogenic gene in placenta, and to check out whether associations between expression and PAEs concentration, to provide evidence for the adverse effect of PAEs to People.Method:To collect urine of90pregnant women before delivery, using questionnaire to investigate the lifestyle during pregnancy, collecting the placenta of pregnant women, to measure the concentration of four PAEs metabolites by SPE-LC/MS method, to analysis lifestyle differences in high PAEs-concentration group and controls using Binary Logistic; to check out the expression of steroidogenic gene in placenta (CYP11A1,P450C17,3beta-HSD) using RT-QPCR; to calculate the correlation of expression of gene and concentration of PAEs using Pearson Correlation analysis.Result:In90pregnant women urine, the detection rate of MMP and the median concentration after creatinine adjustment was98.21%and67.07ug/g respectively; the detection rate of MEP and the median concentration after creatinine adjustment was98.21%and31.97ug/g respectively; the detection rate of MBP and the median concentration after creatinine adjustment was85.71%and16.85ug/g respectively;the detection rate of MEHP and the median concentration after creatinine adjustment was98.21%and47.76ug/g respectively; After binary logistics, house fitment was the influencing factor in high MBP concentration group, we didn’t find any other factor.After extraction of90placenta total RNA, reverse transcription and RT-qPCR,, absolute quantitative method calculate the expression, the median level of three transcript was3betaHSD (6.892×104), CYP11A1(3.825×104), P450C17(1.878×104) respectively; after logit-tansformed; MMP was correlation with CYP11A1, the coefficient of correlation and p value was0.356and0.001respectively; MMP was correlation with3b-HSD, the coefficient of correlation and p value was0.295and0.004respectively MEP was correlation with CYP11A1, the coefficient of correlation and p value was0.249and0.030respectively; after grouped by quintile, quintile3MMP was negative correlation with CYP11Al, the coefficient of correlation and p value was-0.565and0.012respectively, quintile5MBP was correlation with3b-HSD, the coefficient of correlation and p value was0.470and0.042respectively; quintile5MEHP was negative correlation with CYP11A1, the coefficient of correlation and p value was-0.665and0.005respectively; quintile5MEHP was negative correlation with3b-HSD, the coefficient of correlation and p value was-0.524and0.026respectively.Conclusion:The exposure condition of PAEs in Tianjin pregnant women is generally, some PAEs may affect reproduction of people by down-regulation steroidogenic gene expression. |
