Objective: To detect Mdm2and p16and Rb in colorectal cancer tissue and normaltissue differences of Mdm2and p16and Rb in colorectal cancer occurrence anddevelopment of the role and mechanisms of the relationship between the two, for earlydetection, prevention and clinical treatment of colorectal cancer provide a theoreticalbasis.Method:1. Immunohistochemistry staining detected70cases of colorectal cancer and70cases of adjacent non-tumor tissues Mdm2,p16and Rb protein expression, whileanalysis of its relationship with clinicopathological parameters and the two related.2. Reverse transcription-polymerase chain reaction (RT-PCR) detected30cases ofcolorectal carcinoma and30cases of adjacent non-tumor tissues p16mRNA expression,while analysis of its relationship with clinicopathological parameters.Results:1.70cases of colorectal carcinoma in Mdm2protein positive expression rateof67.1%(47/70), positive staining was mainly localized in the cyteblast of tumor cells,showing brown granular distribution of70cases of adjacent noncancerous tissuesMdm2protein positive expression rate of8.57%(6/70), there is significant differencebetween the two groups (P <0.05); colorectal cancer tissues Mdm2protein expression isnot significant difference with sex, age and tumor related (P>0.05), However, with thedegree of differentiation, Dukes of clinical stages, serosal invasion, lymph nodemetastasis (P <0.05).2.70cases of colorectal cancer tissues p16protein positive expression rate of47.14%(33/70), positive staining was mainly localized in the cyteblast or cytoplasm of tumorcells, showing brown granular distribution of70cases of adjacent noncancerous tissuesp16protein The positive expression rate was87.14%(61/70), there is significantdifference obviously (P <0.05). Colorectal cancer tissues p16protein expression is notsignificant difference with sex, age, tumor location (P>0.05), but with the degree ofdifferentiation, Dukes of clinical stages, histological grade,serosal invasion and lymph node metastasis (P <0.05).3.70cases of colorectal cancer tissues Rb protein positive expression rate of34.28%(24/70), positive staining was mainly localized in the cyteblast of tumor cells, showingbrown granular distribution of70cases of adjacent noncancerous tissues Rb protein Thepositive expression rate was84.28%(59/70), there is significant difference obviously (P<0.05). Colorectal cancer tissues Rb protein expression is not significant difference withsex, age, tumor location (P>0.05), but with the degree of differentiation, Dukes ofclinical stages, histological grade, serosal invasion and lymph node metastasis (P<0.05).4.70cases of colorectal carcinoma in the Mdm2protein and Rb protein expression wasnegative correlated (r=-0.264P <0.05).5.70cases of colorectal carcinoma in the Mdm2protein and P16protein expressionwas not correlated (r=1, P>0.05)6. p16mRNA in colorectal cancer tissues than adjacent noncancerous colorectalorganizations (P <0.001). p16mRNA expression and degree of differentiation, Dukesof clinical stages, serosal invasion and lymph node metastasis (P <0.05), with gender,age and tumor related (P>0.05).Conclusion:1. Mdm2expression in colorectal cancer was significantly higher, and the degree ofdifferentiation of colorectal cancer, Dukes of clinical stages, serosal invasion andlymph node metastasis significantly correlated;2. p16expression in colorectal cancer is also significantly lower, and with the Dukesstaging of colorectal cancer, serosal invasion and lymph node metastasis is alsosignificantly correlated,3. Rb expression in colorectal cancer is also significantly lower, and with the Dukesstaging of colorectal cancer, serosal invasion and lymph node metastasis is alsosignificantly correlated,4. Mdm2and Rb a negative correlation between protein expression, indicating bothwith invasion and metastasis of colorectal cancer have a close relationship.5. Mdm2and p16a not correlation between protein expression, indicating both with invasion and metastasis of colorectal cancer have not close relationship.6. Combined determination of Mdm2and p16and Rb may have important clincalsignificance.It would be a good index of predicting prognosis and offering proof fortreatment...
