Objective:Analyze the correlation between the expression of RNF2 and P53/MDM2 in human colorectal cancer tissues and adjacent marginal non-cancerous tissues,and explore their relationship with clinical pathological factors and possible mechanisms in the development of colorectal cancer.Methods:46 patients,including 27 males and 19 females,aged between 26 and 82 years old,with primary colorectal cancer tissues were chosen,which were resected by the Second hospital,University of South China.S-P three-step immunohistochemical staining was used to detect the expression levels of RNF2,P53,MDM2 in samples,and compare the differences of expression level of the colorectal cancer tissues and its adjacent margin non-cancerous tissues,and analyze the possible association of RNF2,P53,MDM2 expression levels with different clinicopathological features(age,sex,tumor size,depth of invasion,presence or absence of lymph node metastasis,presence or absence of distant metastasis,degree of differentiation).Results:(1)RNF2,P53 and MDM2 were expressed in different degrees in human colorectal cancer tissues and adjacent non-cancerous tissues.(2)The positive rate of RNF2 in human colorectal cancer tissues and the positive rate in adjacent marginal non-cancerous tissues was 71.7%and50%respectively.The results comparison between groups showed that expression differences of RNF2 in human colorectal cancer tissues and adjacent marginal non-cancerous tissues was statistically significant(?~2=4.563,p<0.05).RNF2 was related to the presence or absence of lymph node metastasis and TNM staging,and the difference was statistically significant(?~2=5.820,p<0.05).(3)The positive rate of P53 in human colorectal cancer tissue and adjacent marginal non-cancerous tissues was 69.6%and 2.2%respectively.The results comparison between groups showed that the expression differences of P53 in human colorectal cancer tissue and adjacent marginal non-cancerous tissues was statistically significant(?~2=45.409,p<0.05).P53was related to the presence or absence of distant metastasis,and the difference was statistically significant(p<0.05).(4)The positive rate of MDM2 in human colorectal cancer tissue and adjacent marginal non-cancerous tissues was 63%and 32.6%respectively.The results comparison between groups showed that expression differences of MDM2 in human colorectal cancer tissue and adjacent marginal non-cancerous tissues was statistically significant(?~2=8.538,p<0.05).(5)There was no significant correlation between RNF2 protein expression and P53 and MDM2 protein expression(p>0.05).Conclusions:(1)RNF2,P53 and MDM2 proteins are highly expressed in human colorectal cancer,it may be related to the occurrence of colorectal cancer;(2)The expression of RNF2 is related to the presence or absence of lymph node metastasis and TNM staging in colorectal cancer patients,it may have been involved in the process of metastasis and invasion of colorectal cancer;(3)In colorectal cancer,there is no correlation between RNF2 and P53/MDM2 negative-feedback loop.
