Objective: To discuss the effect of small needle knife therapy on fracture healing and expression of TGF-β1in the callus. Provide a theoretical basis and experimental basis for further experimental study and clinical application, and provide new ideas and methods for clinical treatment of fractures.Methods:72SD rats were randomly divided into four groups, the experimental group (A), model group (B), sham operation group (C), normal group (D), n=18. Group A and B were caused fracture model by surgical operation, group C was just incised skin, not causing a fracture. In the experimental stage, group A be treated of small needle knife, the other groups were without any treatment.1,2and3weeks after surgery, six SD rats in each group were killed to get the fracture specimens. To evaluate the fracture healing by gross observation, callus measurement, morphological observation, immunohistochemistry of TGF-β1and analyse them by statistics.Results:1.2and3weeks after surgery, callus thickness of group A was significantly higher than the group B. 2. Morphological observation show that chondrocytes and osteoblasts of group A appeared earlier and more than group B.3. TGF-β1in immunohistochemistry show that2,3weeks after surgery, content of TGF-β1in group A was significantly higher than in group B and group D, which in the mature cartilage cells stained deepest; Group C and Group D are difficult to find positive staining cells.Conclusion:1. The therapy of small needle knife can promote the fracture callus proliferation and promote early fracture healing.2. The therapy of small needle knife can improve the content of TGF-β1in the callus and osteogenesis role of it is related to expression of TGF-β1.
