Objective:To explore the therapeutic effect of Huoluo Xiaoningdan Jiajianfang (HLXLDJJF) on osteoarthritis animal models, then to further explore its potential mechanism of action.Methods:Prepare dry powder of HLXLDJJF crude drug extract, then duplicate rat granuloma model, mouse ear edema model, mouse writhing pain model and osteoarthrits rat models caused by surgical method or enzymatic method respectively. After drug administration, the weight of granuloma, the weight of ear loop and the times of writhing are determined to evaluate its therapeutic effect on these models. After administration to osteoarthritis models, observe changes on pathological slices to evaluate its effect on pathological changes. Pick these serum of osteoarthritis rats induced by surgery, test the content of T-AOC, NOS, SOD, MDA, TNF-α and IL-17A, to explore potential actional mechanisms of this decotion.Results:HLXLDJJF could reduce the chronic inflammation of rat granuloma, reduce the acute inflammation of mouse ear swelling, ease pain of mouse writhing test; and also partly remiss knee’s pathological changes of osteoarthritis rats induced by surgery, and reduce the generation of articular cartilage pannus.Meanwhile, this decotion could enhance the ability of T-AOC in rats induced by surgery, and reduce the production of NO, increase the content of SOD, and decrease the contents of TNF-α and IL-17A in serum.Conclusion:HLXLDJJF has the abilities of anti-inflammatory, blood stasis removing and analgesia, and has the ability of palliate the pathological process of osteoarthritis. Potential action mechanism of this decotion could be enhancing body’s ability of antioxidant, inhibiting inflammation response, so to achieve the role of palliate cartilage damage process.
