Effects Of Toutongning Capsule On The The Expression Of Substance P MRNA In The Midbrain In Migraine Raet

Object：To investigate the effects of Toutongning capsule on the expre-ssion of substance P mRNA in the midbrain in migraine rats.Method： The24adult Wistar rats were divided into4groups.Toutongning capsule treatment group(D) and model group(B), the migraineheadache was induced by subcutaneous injection of nitroglycerin. Toutongningcapsule control group(C) and the control group(A), the rats were injected withthe same dose of peanut oil. In the control group(A) andmodelgroup(B).Saline(2ml/Kg/d) was administered intragastrically for7days prior totreatment with peanut oil. In the Toutongning capsulecontrol group(C) andToutongning capsuletreatment group(D), Toutongning capsule(0.375g/Kg/d)was administered intragastrically for7days prior to treatment withnitroglycerin. Two hours following nitroglycerin injection, the rats wereanesthetized with10%chloral hydrate (0.3ml/100g). Six rats from each groupwere decapitated, and the midbrains were isolated and detected substance Pexpression by real-time quantitative PCR.Result：Real-time quantitative PCR results revealed the model groups(B)exhibited an obviously lower level of substance P mRNA expression thancontrol group (A),Toutongning capsule control group(C) and Toutongningcapsule treatment group(D)(P <0.05); no significant difference in substance PmRNA expression in the rat midbrain between control group (A).Toutongningcapsule control group(C) and Toutongning capsule treatment group(D)(P>0.05).Conclusion：Our data obtained support that Toutongning capsule mayexert the therapeutically beneficial effects by up-regulating substance P mRNA expression in the midbrain.
