| (TTN-Capsule) in migraine through the animal experiment,and to explore the effective mechanism in treating migraine.Methods:TTN-Capsule on the part of Pharmacodynamic study,using to stereo- microscope observed mouse in cerebral pia mater microcirculation experi- ments,applying to heat-stimuli induced pain model measuring mouse analge- sia,taking the experimental model of inflammation which was caused by acetic Mouse peritoneal capillary permeability increased.The acute toxicity and long-term toxicity test of TTN-Capsule were carried out according to herbs pharmaco-toxicological definition.Results:1.In normal Mouse cerebral pia mater microcirculation of the expe- riment,TTN-Capsule(large, medium and small)three dose groups compared with the control group, could increase the crossing points with border for the cerebral pia mater microvessel and improve mouse cerebral pia mater micro- circulation,the large dose group was significant(p<0.01).2.In the light and heat induced pain stimulation experiment of mouse, the large dose group of TTN-Capsule compared with the control group at 15 min after intragastric administration,the pain threshold was increased, and had statistical significance(p<0.05).Compared with the control group after int- ragastric administration 30min and 1h,the pain threshold was increased(p<0.01) in the large dose group.Compared with the control group after intra- gastric administration 30min and 1h,the pain threshold was increased(p<0.01) in the medium dose group, and prolonged the analgesia time.Compared with the control group after intragastric administration 1h,the pain threshold had also increased significantly(p<0.05)in low-dose group.Compared with Chinese patent medicine(Zheng Tianwan)group.The analgesic effect of TTN-Capsule's three groups was strong and longer duration.It was obvious that the large dose group of TTN-Capsule had statistical significance(p<0.05,p<0.01).3.In the experiment of inflammation model in mouse that the acetic acid induced enterocoelia capillary perme ability increased,compared with the control group, the absorbance of large dose group decreased significantly (p<0.01);the absorbance of medium dose and Chinese patent medicine(Zheng Tianwan) groups decreased(p<0.05),it was indicated that large,medium dose groups and Chinese patent medicine(Zheng Tianwan)group all had the anti- inflammatory effect.Compared with Chinese patent medicine(Zheng Tianwan) group,the absorbance of three dose groups all decreased,but no statistical significance.It was indicated that TTN- Capsule and Chinese patent medicine(Zheng TianWan)had the equivalency effect in anti-inflammatory.4.The results of acute toxicity test showed that,the maximum of TTN-Capsule to mouse intragastric administration was 36.8 g/kg(163.04g dried herbs/kg).It was equivalent to 460 times in clinic for patient.There was no acute toxicityin in this dose for mouse intragastric administration.5.TTN-Capsule equivalent to 80,40,20times of the amount in adult (60kg) dose got rats intragastric administration to 90 days and 14 days after the drug withdrawal,no rats died,the general situation of rats weight,food intaking,ECG,blood abnormalitiesetc etc.They had no significant change.It was showed that the recommended clinic dosage had the effect of decreasing TG,but did not have side and toxixity.However,liver function might be some affected for long-term.Conclusions:TTN-Capsule with increasing headache cerebral pia mater coll- ateral circulation,could improve the role of microcirculation.TTN-Capsule had some analgesic and anti-inflammatory effect.TTN-Capsule mouse intragas- tric administration of the maximum of 36.8g/kg(163.04g dried medicinal herb /kg),the equivalent of 460 times for people in this dose,had no significant side and toxixity effects.It was indicated that the recommended clinic dosage had the effect of decreasing TG,but did not have side and toxixity.Howev- er,liver function might be some affected for long-term.
