ObjectiveTo investigate the accommodation role of Toutongning capsule on the dura artery blood flow and CGRP level in peripheral blood of the rat models with migraine. To explore the relationship between migraine, CGRP and dura artery blood flow. To identify the therapeutic action mechanism of Toutongning capsule .MethodsThe 24 male SD rats were randomly divided into 4 groups: sham-operated group , normal saline treatment group, sumatriptan treatment group, Toutongning treatment group. Every rat were given drugs according to weight, Toutongning treatment group: Toutongning (3.2 g/kg) was given by ig once daily for 3 days, sumatriptan treatment group: sumatriptan (30mg/kg) was given by ig once daily for 1 days, sham-operated group: normal saline was given by ig once daily for 3 days. normal saline treatment group: normal saline was given by ig once daily for 3 days. One hour after the last ig, Toutongning treatment group, sumatriptan treatment group, normal saline treatment group were made migraine model, sham-operated group were sham operated. The rat model with migraine was manufactured by electrical stimulation of the trigeminal ganglion. We get 1 ml blood of jugular vein from every rats at three times(one hour before to electrical stimulate the trigeminal ganglion, When stop electrical the trigeminal ganglion immediately, one hour after to electrical stimulate the trigeminal ganglion). The radioimmunoassay method was used to mensurate of CGRP level in rat peripheral blood. The laser Doppler instrument was applied to test rat dura artery blood flow at three times (five minutes before to electrical stimulate the trigeminal ganglion, when the trigeminal ganglion was electrical stimulated, five minutes after to electrical stimulate the trigeminal ganglion).ResultsBefore to electrical stimulate the trigeminal ganglion, CGRP level in peripheral blood in several groups were(72.51±6.58)pg/ml in Toutongning treatment group,(70.09±7.70)pg/ml in normal saline treatment group, (64.91±5.27)pg/ml in sumatriptan treatment group,(71.38±3.19)pg/ml in sham-operated group (before sham-operated). When the trigeminal ganglion was electrical stimulated, CGRP level in peripheral blood in rats of the Toutongning treatment group(98.11±6.16)pg/ml was significantly lower than this of normal saline treatment group(125.92±7.59) pg/ml (P < 0.01), which did not have any significant difference when compared with the sumatriptan treatment group(90.55±3.74)pg/ml (P >0.05). And this was significantly different between normal saline treatment group and sham-operated group(after sham-operated)(72.34±6.13)pg/ml (P < 0.01). Before to electrical stimulated the trigeminal ganglion, the dura artery blood flow in several groups were(142.03±52.20)in Toutongning treatment group, (135.89±49.59)in normal saline treatment group, (131.40±72.74) in sumatriptan treatment group, (198.86±80.28) in sham-operated group (before sham-operated) . When the trigeminal ganglion was electrical stimulated, the dura artery blood flow in rats of the Toutongning treatment group(189.51±42.09)was significantly lower than that of normal saline treatment group(240.53±78.39) (P < 0.01), which did not have any significant difference when compared with the sumatriptan treatment group(167.06±75.15) (P >0.05). And this was significantly different between normal saline treatment group and sham-operated group(after sham-operated) (191.01±79.21) (P < 0.01).ConclusionThe CGRP levels in peripheral blood and the dura blood flow increased during electrical stimulated the trigeminal ganglion. So electrical stimulated the trigeminal ganglion can imitate the two significance pathophysiology process when migraine attacked. Toutongning capsule may significantly inhibit peripheral blood CGRP, and also restraint the rat dura blood flow, so it may effectively for treating migraine.
