This dissertation included three parts:Part one:Literature reviewA review about phytochemical investigation and pharmacological activities of plants belong to Osmundaceae Osmunda Linn.Part two:Experimentation1. Thin-layer chromatography of material medicine and crude drugs slice of Osmunda japonica Thunb.According to the thin layer identification method of medicinal materials of Osmunda japonica Thunb of2010edition of the China pharmacopoeia, identified the different origin medicinal materials and crude drugs slices of Osmunda japonica Thunb.The thin-layer chromatography method of medicinal materials of Osmunda japonica Thunb also apply to crude drugs, slices.2. Microscopical chatacteristics of material medicine and crude drugs slice of Osmunda japonica Thunb.Observed the powder of medicinal materials and crude drugs slices of Osmunda japonica Thunb under the10X eyepiece and40X objective, and summarize the characteristics of microscopical chatacteristics. The characteristics of microscopical chatacteristics are:Powder is brown. Starch grains are nombreux. The shape of the starch grains are round, oval, close to the round of irregular form. Hilum points are humanoid shape or punctiform shape. Diameter of starch grains are5~15μm. Diameter of margin of grain hole shape tracheids are30~45μm. Diameter of tracheids of ladder form are25~50μm. Parenchyma cells are round or long hexagon. Parenchyma cells are not contain calcium oxalate crystallization. The color of wooden fiber cell is depth. The cell walls of wooden fiber cell are thicker.3. Inspection of crude drugs slices of Osmunda japonica Thunb. According to the2010edition of the China pharmacopoeia medicinal materials of Osmunda japonica Thunb inspection standard and extract the standard inspection method for different origin crude drugs slices for inspection, including the moisture content determination, total ash content determination, acid insoluble ash content determination, determination of extracting substances.4. The quantitative determination of osmundacetone by HPLC from Osmunda japonica Thunb.Established the quanlity controlling method of osmundacetone by HPLC from Osmunda japonica Thunb.Regress equation of osmundacetone was: Y=6.3135X-0.8554(r=0.9999), which showed that the calibration curve was linear within the range of4.22ng-63.30ng. The average recovery rate was98.7887%. The result of precision, repeatability and stability all met our requirements, showed that the methods were steady and reliable and can be used as the determination methods. The cotent of osmundacetone in medicinal materials ranged from0.07677mg/g~0.09293mg/g. The cotent of osmundacetone in crude drugs slices ranged from0.06533mg/g~0.00901mg/g.5. The fingerprint of Osmunda japonica Thunb.Established the fingerprint of Osmunda japonica Thunb.The absorbency in320nm. Relative retention time and relative are of single peak were determined. In experiments of accuracy and reproducibility, RSD of relative retention time were0.082%~2.984%and2.320%~4.988%, RSD of relative peak area were0.211%~1.490% and3.468%~5.431%. The stability experiment revealed that the sample solution is stable in21hours. Data were analyzed by "Sop of Similarity evaluation system for chromatographic fingerprint of TMC(Version2004)" software to compare the medicinal materials were all above0.90, and the crude drugs slices were all above0.91.6. The extraction of effective parts and antibacterial activity screeningWith the refluxing method and system solvent extraction method, extracted and isolated the steroids, esters, glycosides and polysaccharides of Osmunda japonica Thunb. The KB paper diffusion method was performed to screen the antibacterial activity of these effective parts,including escherichia coli, staphylococcus aureus, pseudomonas aeruginosa and Candida albicans. The effective parts in10g content/ml dose were not significantly antimicrobial activity.Part three:Summary and discussionThis dissertation makes a foundation for further research of Osmunda japonica Thunb.
