Chromatography fingerprints can comprehensively reflect the whole chemical characteristics and intrinsic quality of the herbs. They also cover the disadvantages of quality control with few effectives in the past. So this is one of more reasonable important model. Its unique advantage obtained increasing numbers accreditation. At present the fingerprint techniques are most effective quality controlling methods of traditional Chinese medicines or herbal medicines. They were very useful for medicinal plant identification, GAP bases establishment of traditional Chinese medicines, prior selection for germplasm resource, origin evaluation of medicinal materials, quality assessment and so on. The present research mainly focuses on the fingerprint establishment of flowers of Lonicera japonica Thunb. and Salvia miltiorrhiza Bge by GC, GC-MS and HPLC, combined with the quantitative analysis multi-target ingredients. The results are as follows:1. The establishment of the GC fingerprint of the essential oils from the flower of Lonicera japonica Thunb.The GC fingerprint was established after the analysis of the essential oils from the flower of Lonicera japonica Thunb. with 10 batches samples by GC method. The capillary gas chromatography was used with hydrogen flame ionization detector, to compare the similarity among the flower of Lonicera japonica Thunb. samples from different origins. After the fully component separation of medicinal materials, 26 peaks were determined as common fingerprint peaks. The GC fingerprint of essential oil of the flower of Lonicera japonica Thunb. can be used as one of its testing standards.2. The establishment of the GC fingerprint of the essential oils from Salvia miltiorrhiza Bge. flowerTo utilize Salvia miltiorrhiza Bge.( S. miltiorrhiza ) resources comprehensively, studying on purple flower and white flower, the essential oils components of S. miltiorrhiza flower of 7 samples from 6 origins were analized and then the GC fingerprint was established. The chemical components in the essential oil of S. miltiorrhiza flower of each origin were analyzed with the GC-MS and 82 compounds were identified from all the samples. The essential oils dominated by the sesquiterpenes, fatty acids, and alkanes, includingβ-Caryophyllene,β-Caryophyllene oxide,α-Caryophyllene, and Cadindiene. Hexadecanoic acid was the main fatty acid component. The highest amount component (12.2-31.7%) in the essential oils of S. miltiorrhiza flower is theβ-caryophyllene with the functions of anti-tumor, anesthetic, antifungal and preventing and curing of colitis disease.3. The establishment of HPLC fingerprint of the flower of Lonicera japonica Thunb.Methanol extracts of the flower of Lonicera japonica Thunb. from 11 different origins were analyzed with HPLC, and 16 common peaks were confirmed by using the gradient elution with methanol-0.1%phosphoric acid water solution as mobile phase under the detection wavelength of 240nm. The contents of chlorogenic acid and lutelin 7-o-glucoside in the flower of Lonicera japonica Thunb. of various origins were also determined,1.46-3.71%, 0.55-2.04% respectively, chlorogenic acid content and lutelin 7-o-glucoside contents accord with that regulated by Chinese Pharmacopoeia. The data was analyzed by the software"Computer Aided Similarity Evaluation"recommended by Chinese Pharmacopoeia Commission. After the similarity analysis of fingerprints and clustering of the flower of Lonicera japonica Thunb. from 11 origins, it was believed that the HPLC fingerprints had good reproducibility. And the established fingerprint of the flower of Lonicera japonica Thunb. can be used for their quality control.4. The establishment of the HPLC fingerprint of S. miltiorrhiza flowerThe S.miltiorrhiza contains the liposoluble and water-soluble components. The ultrasonic extraction method with 70% methanol can effectively extract chemical components of S. miltiorrhiza flower. The establishment of fingerprint chromatograph was obtained from the measurement of 10 different origins. The effective components were analyzed with the seven reference substance, including Danshen su, protocatechualdehyde, caffeic acid, salvianolic acid B, TanshinoneⅠ, dihydrotanshinoneⅠand Tanshinone IIA. The results showed that these components exist except Tanshinone IIA. The preliminary results indicated that S. miltiorrhiza flowers may be taken as raw material of Cardiotonic Pills. By comparison of the effective components in the roots, stems, leaves, flowers, and fibrous root, it can be realized that the contents of salvianolic acid B and TanshinoneⅡA in the fibrous root had higher contents than those in other parts.
