| Osmunda japonica, a medicinal plant, belongs to the family of Osmundaceae, which widely distributed in China. Osmundae rhizoma (Ziqiguanzhong), the rhizome of O. japonica is recorded in(2010 edition) as a herbal drug with the efficacies of removing toxic heat, expelling parasites, and stopping bleeding in terms of TCM. It has been widely used for the prevention and treatment of influenza clinically. The aerial part of O. japonica riches in vitamins, trace elements, proteins and essential amino acids, also has some similar efficacies with the rhizome. In order to take advantage of the resources of O. japonica reasonably, the following researches related its chemical components have been done.1. Nine compounds were isolated from aerial part of O. japonica by solvent a nd chromatographic methods and identified as Dehydrovomifoliol (OSA-1), Os mundacetone(OSA-2),4-Hydroxybenzylideneacetone(OSA-3), Protocatechuicaldeh yde(OSA-4), Protocatechuic acid(OSA-5), p-Hydroxy Benzaldehyde(OSA-6),15-Nonacosanone(OSA-7),β-Sitosterol(OSA-8),β-Daucosterol(OSA-9) by spectral a nalysis. OSA-1, OSA-3, OSA-4, OSA-6, OSA-7 were isolated from the genus osmunda for the first time.2. Osmundacetone was synthesized by condensing protocatechuic aldehyde and acetone in NaOH solution. Then, It was purified by silica gel column eluted w ith petroleum-acetone(7:3). The pure product was tested by HPLC and TLC an d confirmed the purity was higher than 98%.3. Microwave assisted extraction-HPLC/UV was used to detect the content of Osmundacetone in O. japonica. The results show that the content of osmundacetone of O. japonica from six different harvest times was 0.0013-0.0073% in aerial part and 0.037~0.058% in rhizome. The content in the rhizoma is 5-45 times higher than the aerial part overall.4. The method of Posphorus molybdenum-tungsten acid casein was used for determining the content of tannins in O.japonica. The results show that the content of tannins of O. japonica from six different harvest times was 0.40%~2.37% in aerial part and 040%~3.88% in rhizome. The tannins are richer in the rhizome than the aerial part.5. The method of Sulfuric acid-phenol was used for determining the content of polysaccharides in O. japonica. The results show that the content of sugar of O. japonica from from six different harvest times was 1.21%~3.05% in aerial part and 0.97%~1.75% in rhizome. The content of polysaccharides in the aerial part is higher than the rhizome.6. The volatile constituents from aerial part and rhizome of O. japonica Thunb. were analyzed by head-space solid micro-extraction, coupled with GC-MS. Thirty-one compounds were identified from the aerial part of O. japonica and twenty-seven compounds from the rhizome; Eight compounds were owned in both samples, account for 13.99% of total compounds in the aeial part and for 40.04% in the rhizome. The results show that the volatile constituents in aerial part of O. japonica are quite different from that in rhizome. |
