Background and ObjectivesCondylomata acuminatum(CA) is a sexually transmitted disease (STD),and by the human papillomavirus (HPV) infection areas such as the anus,perineum and genitalia caused by a common sexually transmitted diseases. The virus is in squamous epithelium copy of double strands of DNA viruses,through the damaged skin contact with infected spread; In recent years,its incidence is increasing rapidly,according to China’s venereal test reports,genital HPV testing rate was30.69/100,000accounts for our country the genital tract sexually transmitted diseases of second;In a foreign country,its incidence is second only to gonorrhea,so the study of mechanism to control the CA has the very vital significance.This experiment through to the p-STAT3and COX-2in CA tissue the expression and relevance of research,this paper discusses the pathogenesis of Condylomata acuminatum,for its prevention and treatment to provide the theory basis.Rapid growth and easy recurrence were the characteristic of CA,a few of CA due to hyperplasia become giant condyloma,part of occur malignant tumor.In recent years, the studies found that cell apoptosis and proliferation abnormalities in human squamous epithelial cells infected with HPV,more research on its mechanism,but, p-STAT3and COX-2were few of studies,in the pathogenesis of CA and genital cancer. Therefore, we using immunohistochemistry SP were detected in43cases of CA and32cases of normal epithelial tissue,p-STAT3and COX-2expression,and to analyze the relevance of p-STAT3and COX-2expression of CA.Materials and Methods1Materials43cases of patients with CA from the First Affiliated Hospital of Zhengzhou University Dermatology outpatient,with typical skin lesions,and confirmed by histopathology,including31males and12females;ranging in age from17to55(31.42±7.28) years;course of the disease20~108(54.7±13.63) days.Material did not treatment immunotherapy and other diseases.Urology outpatient surgical resection in32cases of normal adult male circumcision as a control.All tissue samples drawn were placed in a poly-formaldehyde solution of40g/L fixed, paraffin-embedded,consecutive4μm sections.2MethodsSP immunohistochemistry method was used to measure the expression of p-STAT3and COX-2in tissue samples from43cases of CA and32normal skin controls.Operation steps according to instructions. p-STAT3positive is light yellow to brown particles in the nucleus;COX-2positive is the light yellow to brown particles, in the cytoplasm and membrane of the cell.According to the literature scoring methods,Semiquantitative score according to the percentage of positive cells and stain intensity.3Statistical analysisUsing SPSS17.0,application of two independent samples rank sum test to compare differences in levels of p-STAT3and COX-2protein expression in CA tissues, differences in application Chi-Square test was used to compare the positive expression rate,application of Pearson columns contact number analysis of expression of two proteins associated,α=0.05significant level of testing. Results1. The expression of p-STAT3in CA tissues and normal epithelial tissue.p-STAT3positive is light yellow to brown particles in the nucleus. In the43cases of CA,28cases are positive expression and p-STAT3positive expression rate is65.12%(28/43).The intensity of positive expression is++~+++,the positive expression of p-STAT3is mainly located in the nuclei in the prickle cell layer and granular layer of keratinocytes. p-STAT3is not expression in normal epithelial tissues, The difference of positive expression rate between groups was statistically significant(x2=30.53,P<0.05),the difference of positive expression intensity between groups was statistically significant (Z=5.575, P<0.001).2. The expression of COX-2in CA tissues and normal epithelial tissue.COX-2positive is the light yellow to brown particles in the cytoplasm and membrane of the cell. In the43cases of CA,32cases are positive expression and p-STAT3positive expression rate is74.42%(32/43).The intensity of positive expression is++~+++,the positive expression of COX-2is mainly located in the upper part of the stratum spinosum and granular layer,in normal epithelial tissue COX-2has only a small amount of expression. The difference of positive expression rate between groups was statistically significant(x2=31.71, P<0.05), the difference of positive expression intensity between groups was statistically significant (Z=5.713, P <0.001).3. Correlation analysis of p-STAT3and COX-2expression in the CA tissues.In43cases of CA tissue,the posistive expression of p-STAT3is28cases and negative is15cases;in COX-2expression,32cases are positive,llcases are negative. Pearson correlation analysis found that43cases of CA p-STAT3and COX-2expression was positively correlated (r=0.577,P<0.001).Conclusions1. There are an overexpression of p-STAT3in CA tissue and the overexpression may be associated with the over-proliferation of CA tissue.2. There are an overexpression of COX-2in CA tissue, suggesting that COX-2 plays an important role in the occurrence and development of the CA.3. p-STAT3and COX-2expression was positively correlated in the CA tissues,they promote the occurrence and development of the CA.
