This study was conducted to determine the pharmacological effects of different dosage of Erchen Decoction Jiawei on the rats with Spleen Hypofunction and Dampness。This study was finished in two parts。In the first part,30healthy SD(BW120±10g) rats were allotted randomly to two treatments which included control team(Ⅰ) and model team(Ⅱ) by the random figure table.Fifteen rats per treatment were feed in15cages。Only one rat was feed in one cage in every treatment.Rats (Ⅱ)used to establish the model of Spleen Hypofunction and Dampness by feeding GanLan (everyday), lard(5ml;everyday) and smoking(two times per day and15min per time)。Compared with control and data of insufficiency of the spleen and phlegm dampness from references, we can ensure if the model established successfully according to indicators measured in our studyIn the second part,60rats were established the model of Spleen Hypofunction and Dampness by the methods in the first part,then they were allotted randomly in four treatments, Ⅱ、Ⅲ、Ⅳ、Ⅴ treatment respectively.15rats which used as healthy control(I) were feed rat diet during establishing the model。Every treatment were given2ml liguid but II treatment。I treatment were given2ml physiological saline solution, Ⅲ、Ⅳ、Ⅴ were given Erchen Decoction Jiawei respectively6.8g/kg、3.4g/kg、1.7g/kg on day0、1、2、3、4.Five rats of each treatment were slaughtered for collecting lung tissue and serum.Reagent kits were used to determine the expression of SP-A,SP-B in lung and serum SOD,MDA.HE was used in determining the pathological change of lung in each treatment.The results(1) In the first part,compared with control,the rats of model of Spleen Hypofunction and Dampness had extrem significantly decreased in body weight,the positive area of SP-A and average optical density value(P<0.01).The rats of model treatment also decreased in gastrointestinal propellant rate (p<0.05),swimming time (p<0.05),serum SOD (p<0.05) increased in serum MDA (p<0.05)。In addition,the lung of rats in this model existed pathological change of respiratory tract which included ecchymosis, pulmonary alveoli fusion and red seriflux exudation in lung.(2) In the second part,on day9, rats of Ⅰ treatment had normal lung tissue,but Ⅱ existed in disorder lung tissue, broadening pulmonary alveoli mesenchyme, red seriflux exudation in pulmonary alveoli lumen,imperfect pulmonary alveoli.With given Erchen Decoction Jiawei, Ⅲ、Ⅳ、Ⅴ had significant remission effects on pathological change in lung tissue. Ⅳ had the best effects on remission pathological change.(3) On9d,compared with Ⅰ treatment,rats of Ⅱ treatment had decreased in positive area of SP-A, SP-B and average optical density (P<0.05).Compared with Ⅱ treatment,rats of Ⅲ、Ⅳ、Ⅴ treatment had increased in A, SP-B and average optical density (P>0.05), IV treatment had best effects on these indicators and significantly higher than Ⅱ treatment.(4) Compared with0d,on d1、3、5、7, serum SOD of Ⅲ、Ⅳ、Ⅴ treatment increased gradually, and was higher than0、1d。In all time point, Ⅳ treatment which was proximate to I treatment had higher value in SOD thanⅢ、Ⅴ treatment (P>0.05)。 II treatment had lowest SOD value in all time point but on day0, decreased with experiment time。When on day7, SOD value of II treatment was significantly lower than day0、1(P<0.05).(5) Compared with0d,on d1、3、5、7, serum MDA of Ⅲ、Ⅳ、Ⅴ treatment decreased gradually, and was lower than0、1d (P>0.05)。On day7, serum MDA of Ⅳ was lower than day0. In all time point, IV treatment which was proximate to I treatment had lower value in MDA thanⅢ、Ⅴtreatment (P>0.05)。Conclusions:(1) Feeding GanLan,lard and smoking were effective way to establish the model of Spleen Hypofunction and Dampness in rats.(2) Erchen Decoction Jiawei had therapeutical effects on spleen hypofunction and dampness by increasing the expression of SP-A, SP-B in lung, and serum SOD, decreasing serum MDA。 (3) The mechanism of antioxidation and increasing the expression of SP-A, SP-B had positive correlated with pharmacological effects of drying dampness and dispersing phlegm, Qi-regulation of Erchen Decoction Jiawei。...
