Dermatophytosis is a common infectious disease in dogs and cats, and it can be infected each other within pets or between pets and human beings. Recently with the rapid development of medicine technology and resistance of fungi,clinical relapse rate also increased. At the same time, a large number of antifungal drugs were applied in clinical,resistant fungis had been becomed advantages, inducing gene mutations,so that the rate of drug resistance were increased,induced the treatment difficultly. To therapy dermatophytosis,in clinical combination of two antifungal agents has been becomed the research hotspot in recent years, and most effective strategies to solve this problem.The checkerboard microdilution method which according to M27-A2method of CLSI,was applied to study in vitro interactions of four antifungal drugs (terbinafine,5-Flurocytosine,amphotericin B,itraconazole).Pairwise combination of four antifungal and Single agent were applied to treat10stains fungi(mainly were the filamentous fungi and yeast) and made comparison of antifungal activity.Results1:When amphotericin B and5-FC were given in combination,synergy or additive were observed in60%(6/10) of the isolates and40%(4/10) were indifferent,while antagonism was not observed.When amphotericin B and itraconazole were given in combination,synergy or additive were observed in40%(4/10) of the isolates and50%(5/10) were indifferent,10%(1/10) were antagonism.When amphotericin B and terbinafine were given in combination,synergy or additive were observed in60%(6/10) of the isolates and30%(3/10) were indifferent,10%(1/10) were antagonism.When itraconazole and5-FC were given in combination,synergy or additive were observed in60%(6/10) of the isolates and30%(3/10) were indifferent,10%(1/10) were antagonism.When terbinafine and5-FC were given in combination,synergy or additive were observed in50%(5/10) of the isolates and30%(3/10) were indifferent,20%(2/10) were antagonism.When itraconazole and terbinafine were given in combination, synergy or additive were observed in70%(7/10) of the isolates and30%(3/10) were indifferent,while antagonism was not observed.Established dermatophytosis animal model by Microsporum and Epidermophyton floccosum,to observe6groups drugs and "shufu"(ketoconazole) treating dermatophytosis effect.6groups drugs have best ratio,which are AmB+5-FC, AmB+TBNF, AmB+ITRA, TBNF+5-FC, ITRA+5-FC, ITRA+TBNF."shufu"(ketoconazole) is the commonly used antifungal drugs in pet hospital.Results2:The discovery of Microsporum AmB+5-FC, TBNF+5-FC formula to restore the best dog skin damage lesions for (0.5±0.5), To Epidermophyton floccosum AmB+5-FC, AmB+TBNF formula significantly better than the other four formula, lesions for (0.5±0.5), slightly better than "shufu". It shows by AmB+5-FC formula for best compatibility. For clinical combination provides scientific basis.From reported in the literature we can find some trial conclusions which seems to be self-contradictory. For example, in vitro antifungal sensitivity the same combination on different fungi have different interactions,and sometimes even the same species have different interaction.But in the clinical case reports found thatin small animal who has same disease,using the same combination are also different effets.Results3:TBNF+5-FC:3cases were cured, AmB+5-FC:4cases were cured,2cases improved,1death. TBNF+AmB:2cases were cured,1improved. A total of nine cases were cured,3cases were improved and1case of death.Add up to,9cases were cured,3cases improved,1death.
