Study On The Bacteriostatic Action And Clinical Effect Of Chuanhuang Powder Against ESBLs-producing E.coli

The aim of this study is to screen the TCM which could eliminate the b acterial resistance(or enhance the antibacterial effect of the antibiotics), then make up prescription and evaluate the effect. Meawhile, the clinical applicatio n effect and the mechanism of antibacterial was studied by animal efficacy te st and the mechanism study, this could provide basis for rational drug use.1. The minimum inhibitory concentration(MIC) of common antibiotics wa s determined and ESBLs-producing was detected. The gene subtype of ESBLs-producing were determined by PCR. The results showed that resistance rate o f strains to Ceftriaxone, Ceftazidime, Cefotaxime, Amoxicillin et al as high as100%; All of the strains contained the drug resistance gene; Six strains cont ained two genotypes, Three strains contained three genotypes.2. The influence of Chinese herbal medicine on the MIC of drug resistant ba cteria was determined by the passage of the combination of TCM and Wester n Medicine(WM) and screened the TCM which could inhibit bacteria action,th en we suggested that Chuanghuang Powder was composed of Andrographis pa niculata, Scutellaria baicalensis Georgi, Dendranthema indicum and then detect ed the changes of MIC in the same way.The result showed that Chuanghuang Powder could increase the antimicrobial activity of Cephalosporin Quinolones et al.3 10 days San Huang chicken were selected to design the challenge test in v ivo with drug resistance strains and conducted the treatment test with the Diff erent doses of Chuanghuang Powder and Bailong Powde water extracts for th e diseased chicken flocks.The results showed the cure rate of the high dose group and medium dosage group of Chuanghuang Powder was 86.7%、83.3% r espectively, but the high dose group and medium dosage group of Bailong Po wder was just 70% and 56.7%, It suggested that the effect of Chuanghuang P owder was better than Bailong Powder.4.The effects of Chuanghuang Powder water extract on ultrastructure of cell wall were explored, including of the change of AKP content in bacteria liquid, cell membrane permeability, the extracellular soluble protein content, the total protein content, the change in ultraviolet absorption of bacteria and the E.coli DNA. And this experiment clarified the antibacterial mechanism of Chuanghuang Powder water extracts on E.coli were that this could improve the contents of extravasation by increasing the cell wall and cell membrane permeability and inhibit bacterial DNA replication and bacterial protein synthesis.5. The study showed that clinical treatment of Chicken E.coli disease effect is significant, the average cure rate was 96.3%, and the effective rate was 91.7%.
