[Purpose]By adding components of natural drug such as icariin, berberine and ligustrazine respectively to the culture fluid of Yanbian cattle’s ear fibroblasts in vitro respectively, this experiment was to test their influence on the vitality and oxidation repair of these fibroblasts, thus to determine the optimal concentration of the components of natural drug for improving the vitality of Yanbian cattle’s ear fibroblasts and to evaluate their ability to repair the oxidative demage of cells. This experiment will provide technician support to the better application of Yanbian cattle’s ear fibroblasts in semiotic cell cloning and the work of conserving breeds for Yanbian cattle in the future.[Methods]1. After having cultured the Yanbian cattle ear fibroblasts in vitro to the fourth generation, add icariin, berberine and ligustrazine of different concentration to the DMEN respectively. After24,48,72,96hours, use MTT methods to test the vitality of the fibroblasts, analyze all the data collected and then single out the optimal concentration.2. Add two of these three components, each of their optimal concentration, at the same time to the DMEN, after96hours, use MTT methods to test the vitality of the fibroblasts and analyze the all the data collected.3. Add icariin, berberine and ligustrazine, each of their optimal concentration, to the Yanbian cattle ear fibroblasts bearing oxidative damage respectively, after96hours, use MTT methods to test the vitality of the fibroblasts, analyze all the data collected, compare with the control group and the self-repair group, and then evaluate their ability to repair the oxidative damage. [Results]1. In the experiment to test icariin’s influence on cattle ear fibroblasts, all the icariin test group can improve the vitality of the Yanbian cattle ear fibroblasts. The vitality of the fibroblasts with icariin of concentration from0.001μg/ml to0.1μg/ml is significantly higher than that of the control group after72hours of culturing, and when the concentration is0.001μg/ml, it has the best effects.2. In the experiment to test berberine’s influence on cattle ear fibroblasts, the concentration from0.01μmol/L to1μmol/L can improve the vitality of the Yanbian cattle ear fibroblasts. The vitality of the fibroblasts with berberine of concentration from0.01μmol/L to1μmol/L is significantly higher than that of the control group after72hours of culturing, and when the concentration is1μmol/L, it has the best effects.3. In the experiment to test ligustrazine’s influence on cattle ear fibroblasts, the concentration from25μg/ml to50μg/ml can improve the vitality of the Yanbian cattle ear fibroblasts. When the concentration is50μg/ml, it has the best effects and the vitality of the fibroblasts is significantly higher than that of the control group after72hours of culturing.4. In the experiment to test the influence of the combined components on Yanbian cattle ear fibroblasts, the berberine&ligustrazine group and the icariin&berberine group restrain the vitality of Yanbian cattle ear fibroblasts, while the icariin&ligustrazine group improves the vitality of the fibroblasts, though there is no obvious difference between this group and the control group.5.1n the experiment to test the ability of icariin to repair the oxidative damage of Yanbian cattle ear fibroblasts, the fibroblast vitality of the repair group is significantly higher than that of the self-repair group, while is insignificantly lower than that of the control group. The results indicate that icariin has the ability to repair oxidative damage.6. In the experiment to test the ability of berberine to repair the oxidative damage of Yanbian cattle ear fibroblasts, the fibroblast vitality of the repair group is significantly higher than that of the self-repair group, while is significantly lower than that of the control group. The results indicate that berberine has the ability to repair oxidative damage.7. In the experiment to test the ability of ligustrazine to repair the oxidative damage of Yanbian cattle ear fibroblasts, the fibroblast vitality of the repair group is significantly higher than that of the self-repair group, while is significantly lower than that of the control group. The results indicate that ligustrazine has the ability to repair oxidative damage.
