| The research object ot this project was Chimonanthus Praecox seeds as, the results are shown as below:(1) The Solvent Extraction Method was studied by making use of single factor experiments and the orthogonal experimental design. The results showed that the optimal extraction technology conditions for Chimonanthus praecox seed oil were as follows:using the petroleum ether as the solvent, the ratio of material to liquid1:8, soaking time75min, extract2times. In this way, the oil yield was34.0%.(2) The physical and chemical characteristics of Chimonanthus Praecox seeds oil was investigated by using of methods in relevant state standard and appendix VI and appendix VII in "Chinese pharmacopoeia (2010)", and the relative percentage of saturated and unsaturated fatty acids in the seeds oil was measured by GC-MS. Results showed that total content of the unsaturated fatty acid both seed oils was above80%in both kinds of seeds oil which was made by solvent extraction method and expelling method. Both of them were non-drying oils with high value in development and application prospect.(3) The alkaloids in Chimonanthus Praeocox seeds was studied by extraction, separation, purification and structure identification.Firslty, the silica gel column chromatography and recrystallization method was used in the separation and purification of alkaloids.Then, the purity of alkaloid monomer was qualitative analysed by using of the thin layer chromatography (TLC) and high performance liquid chromatography (HPLC). Results showed that this reseaech got a higher purity of alkaloid monomer. Accord ing to the analysis of alkaloids related nuclear magnetic resonance (NMR) specturms, the alkaloid monomer was Chimonanthine. |
