Fluorescence detection method of embedded fiber optic has been a means of capillary electrophoresis chip detecting, with simple optical structure and small size and without complex optical instruments, and with incomparable advantages to the conventional confocal detection, and it had an important role in developing detecting system miniaturization and integration. Based on ARM9 processor and embedded Windows CE operating system, laser-induced fluorescence detection system of embedded fiber-optic without computer was designed to achieve the functions of fluorescence signal acquisition and chip's image observation of separation process.Detection system of Optical Fiber Embedded Electrophoresis Microchip mainly includes system's optical design, detecting hardware platform, the upper application and production of CE chip. Based on the optimization of fiber-optic detection parameters, CE chips with different structures were made by MEMS technology.The system used PMT to achieve photoelectric conversion, and the signal would be amplified and filtered and digital-analog converted, PMT sensitivity control circuit was designed to improve detection range of the system. Based on the ARM9 hardware platform, the CMOS camera OV9650 was used to acquire images which displayed on the LCD screen.CE_LIF applications called the drive of bottom circuit and achieve real-time displaying data.Using the detection system designed, the images of injection and separation of the sample were observed, which could help analyze the whole process of CE. CE separation experiments were made for different concentrations of Rhodamine B samples with the detection system, and the minimum detection limit of the system is 1×10-6mol/L. The experiment results showed that the design of optical fiber embedded capillary electrophoresis with fluorescence detection system had a high detective sensitivity and enables visualization of the analysis process.
