Psoriasis vulgaris is most comme type.Psoriasis vulgaris isdivided into different stages by the course of disease.The diagnosis of thisdisease is easy,but the pathogenesis and the treatment is a large problem totrouble the doctors.Past research showed psoriasis is a T lymphocyteabnormality,Th1cells with excessive activation for advantage leads to theautoimmune diseases.Recently,in auto-immune disease and allergens specific-ity reaction found a special help type T cells different from Th1cells and Th2cells, called Th17cells,these cells except can specificily secrete IL-17stillsecrete IL-22, IL-21and IL-6etc. It plays an important role in immuneadjustment of autoimmune disease. When the Th17cells found, many scholarsbegan to study its function actively in autoimmune disease. As a commondisease of dermatology,The study of psoriasis vulgaris is a focus todiscuss,more about the study of Th17cells and its effect factors in Psoriasisvulgaris becomes a hot point.We detect the expression level of Th17in peripheral blood andinterleukin-17(IL-17) in blood serum of psoriasis vulgaris,compared withhealthy control volunteers, In order to research the effects of Th17cell and thesecretion of interleukin17with psoriasis vulgaris onset association and theseverity of diease, further discussing the pathogenesis of psoriasis vulgaris. For clinical treatments with new ideas,not only has the academic value butalso has application prosepect.Methods:The case group involved23typical vulgaris psoriasis patientswho were choosed from March2011to December2011in dermatologistout-patient service of the second hospital of Hebei Medical University. Selectstandard:1The case group had typical clinical manifestations of psoriasisvulgaris.2All the cases had not taken steroid and immunosuppressanttherapy in one month.3Excluded patients with autoimmune disease andpatients with other systemic diseases. the case group consist of12males and11females, age range19to53years old, average age was33.30±11.30yearsold.We divided all the cases into two groups,one group was progressive stagewhich had12examples,the other group was stationary stage which had11examples. PASI score which was based on clinical manifestations wasassessed by the same staff.11cases who were from our health medical staffwas normal control group, the group consisted of5males and6females, agerange20to52years old, average age was27.27±8.63years old,Throught-test,sex and age had no significant differences with the case group.1Detect the expression level of Th17cells in peripheral blood:Adopted elbow vein blood4ml from23patients and11normal examplesfor heparin anticoagulation tube,separated the peripheral blood single nuclearcells(PBMC) in8hours,and followed by stimulation with PMA, Ionomycin,Monensinsolution for4hours in37degrees5%carbon dioxide couveuse,Then fixed rupture of membranes, surface and intracellular cytokinestaining,at last determined Th17(CD4+IL-17+T) cells in peripheral bloodlymphocytes by flow cytometry. We analyzed the difference betweent the casegroup and the normal control group,the progressive stage group and thestationary stage group. We also analyzed the relation between the expressedlevel of Th17cells in peripheral blood and the clinical lesions area andseverity degree (PASI score) by spearman rank.2Detect the expression level of IL-17: Adopted elbow vein blood2ml from23patients and11normal examplesfor anticoagulation tube with10%EDTA centrifugated samples.Then theserum samples were frozen at-20℃in the refrigerator of preservation.Application of double antibody sandwich-type enzyme-linked immunosorbentassay (ELISA) detected the expression level of IL-17in blood serum. Weanalyzed the difference betweent the case group and the normal control group,the progressive stage group and the stationary stage group.We also alyzed therelation between the expressed level of IL-17in serum and the clinical lesionsarea and severity degree (PASI score) by spearman rank.Result:1The result of Flow Cytometry was showed the expression level of Th17cells in psoriasis vulgaris peripheral blood (2.09±1.53)%was higher than thatin the normal control group (0.96±1.02)%(t=2.22P=0.03P<0.05), thedifference was statistically significant; the difference between the progressionstage group (2.14±1.72)%and the stationary stage group (2.04±1.39)%was nostatistically significant (t=0.16P=0.87P>0.05); the expression level of Th17cell and the clinical area and severity degree (PASI score) had not linearcorrelation(r=-0.22P=0.31P>0.05).2The result of enzyme-linked immunosorbent assay was showed, theexpression level of IL-17in psoriasis vulgaris serum30.85±11.52was higherthan that in the normal control group21.87±9.99(t=2.21P=0.03P<0.05),thedifference was statistically significant; the difference between the progressionstage group32.07±10.75and the stationary stage group29.51±12.69was nostatistically significant (t=0.52P=0.61P>0.05);the expression level of IL-17and the clinical area and severity degree (PASI score) had not linearcorrelation(r=0.27P=0.22P>0.05).Conclusion:1The expression level of Th17cells in peripheral blood of psoriasisvulgaris group was higher than that in the normal control group, explained thatTh17cells may participated in the process of psoriasis vulgaris, directly confirmed the expression of Th17cells may play a important role in thepathogenesis of psoriasis vulgaris.2The expression level of IL-17in the serum of psoriasis vulgaris groupwas higher than that in the normal control group, explained that IL-17who iseffectors of Th17cells may participated in the process of psoriasis vulgaris,indirectly confirmed the expression of Th17cells may has correlation with thepathogenesis of psoriasis vulgaris.3the expression of Th17cells in peripheral blood and IL-17in the serumof psoriasis vulgaris in the stage group and that in the stationary stage groupwas no different, indicated Th17cells and IL-17may play an importan role inthe whole flammatory progresses of psoriasis lesions, the measured value ofTh17cells and IL-17can not reflect the period of this disease.4The expression level of Th17cells in peripheral blood and IL-17in theblood serum of psoriasis vulgaris had not correlation with the psoriasis lesionsaera and clinical severity degree PASI score, explained no specific relevancewith the expression level of Th17cells and IL-17the progress of psoriasislesions and severity, so they are not as the evaluation severity index of thedisease. | Keywords/Search Tags: | Psoriasis vulgaris, Th17cells, FCM, IL-17, ELISA | PDF Full Text Request | Related items |
