Objective:To study the relationship between the transcription activity of RORC2protein and the pathogenesis of psoriasis vulgaris of blood-heat type and to investigate the possible role of transcription factor RORC2in the pathogenesis of psoriasis vulgaris of blood-heat type.Methods:By Electrophoretic Mobility Shift Assay (EMSA), the RORC2transcription activity in peripheral blood mononuclear cells (PBMCs) and lesions of patients with psoriasis vulgaris of blood-heat type were detected. The transcriptionactivities in PBMCs of patients with psoriasis vulgaris at different stages were detected. The PBMCs and skin tissues of normal individuals were analyzed as controls. The RORC2transcription activities in PBMCs of six cases with psoriasis vulgaris of blood-heat type before and after the treatment with Liangxue Jiedu Decoction for six weeks were detected also. The comparison between two groups was analyzed by t-test. One-way ANOVA analysis was used for those comparisons among more than three groups. The changes of body surface area and PASI score before and after the treatment were analyzed by Wilcoxon signed-rank test.Results:1、The RORC2transcription activities in PBMCs of25patients with psoriasis vulgaris of blood-heat type were increased obviously compared with those of26normal cases and the difference was statistically significant.2、 There were no correlations between the RORC2transcription activities in PBMCs of patients with psoriasis peripheral of blood-heat type and their body surface area and PASI scores;3、 The RORC2transcription activities in PBMCs of patients at progressive stage were significantly higher than those of patients at stable and receding stages and normal controls. But no significant difference were found between those of patients at stable and receding stage.There were also no significant difference between those of patients at stable and receding stages and normal controls.4、 The RORC2transcription activities in skin lesions of12cases with psoriasis vulgaris of blood-heat type were significantly higher than those of11normal skin tissue and the difference was statistically significant. 5、 The RORC2transcription activities in PBMCs. body surface area and PASI scores were all significantly decreased after the treatment in comparation with those before treatment and the differences were statistically significant.Conclusion:RORC2transcription factor might play a key role in the pathogenesis of psoriasis vulgaris of blood-heat type. It might be an important material base of this type psoriasis vulgaris. Liangxue Jiedu Decoction might work by inhibiting RORC2transcription activity.
