Objective:To study effect of ovary-protecting decoction on ovarian morphology and E2level of rats with controlled superovulation, as well as the effect in different dosage.It provides the theoretical evidence for TCM adjunctive therapy of IVF-ET.Methods:32SPF female rats of8weeks were chose as research object, and were randomized into4groups, modeling group, high-dose medicinal group, moderate-dose medicinal group, low-dose medicinal group,each group had ten rats.The four groups were given intraperitoneal Injection of Alarelin and simultaneously all medicinal groups were given intragastric administration of high, moderate and low dose Chinese medicinals respectively.Till the9th day, HMG was injected to the four groups.HCG was injected after48hours. Then all rats were executed by neck break in batches, at4hours after HCG injection, venous blood were withdrawn. Then, abdominal cavity was opened to take ovarian.Results:1. Serum E2level in moderate-dose medicinal group was higher than modeling group in significant difference (P<0.01) at the day of HCG injectiono Serum E2level in high-dose medicinal group was also higher than modeling group in difference (P<0.05). There is no difference (P>0.05) between low-dose medicinal group and modeling group in Serum E2level.2.Organ index in moderate-dose medicinal group was higher than modeling group in significant difference(P<0.01). Compared with modeling group,Organ index in low-dose and high-dose medicinal group had no difference (P>0.05).3. Observed under light microscope, follicle total number of the ovarian cortex in modeling group were less than all the traditional Chinese medicine group. In particular, the mature follicles was significantly less than all the traditional Chinese medicine group, more atretic follicles.4. Quality follicle rate in low-dose and high-dose medicinal group was higher than modeling group in difference (P<0.05). There is no difference (.P>0.05)between low-dose medicinal group and modeling group in quality follicle rate.Conclusion:1. Ovary-protecting decoction can increase E2level of rats with controlled superovulation at the day of HCG injection.2. Ovary-protecting decoction can promote the development of ovaries and the quality rate of follicle.3. Optimization of E2level, ovarian development and follicle growth by ovary-protecting decoction has certain dose-effect relationship with medicinal concentration.
