ObjectiveAs a mature technology for the treatment of infertility, in vitro fertilization andembryo transfer has been widely used in clinical, but the actual clinical pregnancy rate wasonly20%to30%. Successful IVF-ET requires two important factors, they are High-qualityembryos and Successful implantation of the embryo, Endometrial receptivity is animportant factor in determining the success of embryo implantation. At present, It has beenrelatively easy to obtain good quality embryos, but the impact of endometrial receptivityfactors and the regulation mechanism is very complex, There are a lot of material related toendometrial receptivity. Therefore investigate the mechanism of reduced endometrialreceptivity in the application of ovulation induction drugs and Looking for ways toeffectively combat,It will has great significance in the improving the clinical pregnancyrate of IVF-ET.To explore the effects of ovulation simulants on mouse endometrium andembryo nidation and development so as to provide experimental basis for clinical drugadministration.To investigate the expression of Connexin43(Cx43) in endometrium aftersuperovulation and to explore its role in the depression of endometrial receptivity. And soas to provide experimental basis for raising clinical pregnancy rate of in vitro fertilizationand embryo transfer(IVF-ET).Methods160female Kunming mice were randomly divided into two large groups (pregnancygroup, and non-pregnant group, Each group consisted of80mice), each group was furtherdivided into four groups (a control group and three super-ovulation group), Each subgroupconsisted of20mice,the clomiphene group (group I), the simple human menopausalgonadotropin group (group II), and the combined group consisting of the HMG group(group III). Only to give the saline control group as a control, ovulation-stimulation drugswere given to the The corresponding experimental groups. Respectively, the non-pregnantmice were sacrificed at24h and48h after treatment, and then remove their uteri which were then made into paraffin sections for observation of endometrial development., thenslice HE and immunohistochemical staining, the control group and experimental group, theendometrial development and Connexin43expression in the strength and location, to becompared and analyzed by image processing techniques and strept avidin-biotin complex(SABC) immunohistochemical method. Pregnancy group, the same grouping andadministration, were killed in non-pregnant group of materials in the pregnancy group, themice become pregnant. The females were killed at the7th day, the17d after conception,to observe the differences in all cases of female implantation of fertilized eggs and embryodevelopment of pregnant rats.ResultsNon-pregnant group:(1) endometrial thickness: there is no significant difference among3ovarianhyperstimulation group,(p>0.05), but the thickness of the three super-ovulation group wassignificantly thinner than the control group (p <0.05);(2) gland development:3ovarian hyperstimulation group of GnRHa+simpleovulation group slightly better than that promote ovulation group and the clomiphenegroup, but no statistical significance (p>0.05).(3) connexin43expression:①connexin43expression in the control group aftertreatment,24h,48h was significantly lower than the three experimental groups (p <0.05);②connexin43expression in the clomiphene phenol group was significantly higher thansimply promoting ovulation group and of GnRHa with simply promote ovulation group(p<0.05); connexin43of expression between simply promoting ovulation group and ofGnRHa with simply promote ovulation group no significant significant difference (p>0.05);③connexin43expression in the estrous cycle in time there is a significantdifference (p <0.05).Pregnancy group:(1)embryo implantation rate:3superovulation group was significantly lower than thecontrol group (p<0.05), but there is no significant differences between the threesuperovulation group (p>0.05);(2)differences between the embryonic development: There is no statisticalsignificance between the control group and experimental group (p>0.05).Conclusion1.The development of endometrial impact: GnRHa simple ovulation induction group ofthree experimental groups with minimal impact, endometrial stromal synchronization is the best, the implantation rate is relatively high;2. Embryonic development: embryonic growth was no significant difference betweencontrol group and experimental group, teratogenic rate, stillbirth rate and embryoresorption rate was also no significant difference;3.Connexin43expression: there exists significant difference in terms of time,Its highexpression of endometrial hyperstimulation may closely related to the decrease ofendometrial receptivity after superovulation.
