Partâ… Murine dendritic cells in endometrium and peripheral blood during peri-implantation.Object: To investigate the role of dendritic cells in maternal-fetal immune tolerance,the relative number and maturity of murine dendritic cells in endometrium and peripheral blood during peri-implantation were analyzed.Methord: Flow cytometry and immunofluoresent technique were used to detect the variety of relative number of CD11c~+MHC-â…¡-,CD11c~+MHC-â…¡~+,CD11c~+ endometrium cells and peripheral blood cells during mice peri-implantation.Result: Compared with virgin mice,the relative number of CD11c~+MHC-â…¡-,CD11c~+MHC-â…¡~+,CD11c~+ cells in peripheral blood increased significantly on gestation day 3.5 and 5.5(p<0.05);The percentage of MHC-â…¡coexpression in CD11c~+ cells increased gradually. In the endometrium, there was no significant difference between the relative number of CD11c~+MHC-â…¡-,CD11c~+MHC-â…¡~+, CD11c~+ cells and the percentage of MHC-â…¡coexpression in CD11c~+ cells between virgin mice and those on gestation day 3.5(p>0.05). The increase of relative number of CD11c~+MHC-â…¡- and CD11c~+ endometrium cells of mouse on gestation day 5.5 was more significant than in virgin mouse(p<0.05). The percentage of MHC-â…¡coexpression of CD11c~+ endometrium cells decreased significantly from virgin to gestation day 5.5 mice (p<0.05).Conclusion: The variety of dendritic cells during peri-implantation, espeically the CD11c~+ dendritic cells'increasing, suggested that dendritic cells might candidate on the maternal-feto immune tolerance. Since the variety of dendritic cells in peripheral blood didn't synchronize with endometrium, the peripheral blood couldn't be used instead of endometrium in studies about maternal-feto immune tolerance.Partâ…¡The effect of superovulation on endometrium dendritic cellsObject: To investigate the effect of superovulation on endometrium dendritic cells by analyse the relative number and maturity of murine dendritic cells in endometrium after superovulation.Methord: Flow cytometry and immunofluoresent technique were used to detect the varity of relative number of CD11c~+MHC-â…¡-,CD11c~+MHC-â…¡~+,CD11c~+ decidua cells during mice peri-implantation after superovulation.Result: Compared with spontaneous pregnant mice , the relative number of CD11c~+MHC-â…¡-,CD11c~+MHC-â…¡~+,CD11c~+ endometrium cells on gestation day 3.5 and 5.5 after superovulation increased significantly (p<0.05).The percentage of MHC-â…¡coexpression of CD11c~+ endometrium cells of gestation day 5.5 mice after superovulation was much higher than spontaneous pregnant mice.Conclusion: The effect of superovulation on the relative number and maturity of endometrium dendritic cells perhaps related to the relatively low implantation rate after superovulation.
