Objective:Establish the animal model of acute soft tissue injury,to observe the soft tissue changes of acute soft tissue injury and effects with different cryotheropies to acute soft tissue injury, and to explore possible mechanisms for the clinical treatment of acute soft tissue injury and sports Rehabilitation methods appropriate choice of cold therapy to provide guidance and a theoretical basis.Methods:40 postnatal Wistar rats were usually randomized to blank group(BG),model group (MG),Intermittent cryotherapy group (IG) and continuous cryotherapy group(CG), each group had 30 rats. Established the animal models of Acute Damage of Soft Tissue to the last three groups, hit the inner thigh from the knee 1mm of rats with self-made blowing hammer, in MG no further treatment after injury,then In IG, the rats were treated by Intermittent cryotherapy on the wounded part, and in CG the rats were treated by continuous cryotherapy.48 hours later,all rats were executed, observated the change roughly of the wounded part, and taken the muscular tissue of damage spot.Histological sections were prepared with HE staining for observation of histological change, from the muscle fiber arrangement's disorder, the muscle fiber's breaking, the muscle fiber's swelling, the septum intermusculare's broadening, the phlogocyte infiltration, the red blood cell's seeping out,the muscle fiber denaturedly and textile fiber connection 8 aspects to carry on the histological grading of damage.The expression of IL-1βand TGF-β1 was detected with immunohistochemistry staining. expression was detected by application of tissue superoxide dismutase (SOD) and malondialdehyde (MDA) expression.Results:①The live score of 48 hours later, continuous cold therapy group and intermittent cold treatment group scores were lower than the model group, the results were statistically significant (P<0.05), continuous cold therapy group was lower than intermittent cold therapy group, the results were statistically significant (P<0.05)②histological scores, continuous cold therapy group and intermittent cold treatment group scores were lower than the model group, the results were statistically significant (P<0.05), continuous cold therapy group was lower than intermittent cold therapy group, the results are statistically significance (P<0.05).③Immunohistochemical detection of IL-1βand TGF-β1 expression, compared with the model, continuous cold therapy group and intermittent cold therapy group decreased expression of IL-1β, TGF-β1 expression was increased, the result was statistically significant (P< 0.05), compared with intermittent cold therapy, continuous cold therapy group and the group continued intermittent cold therapy cold therapy group decreased expression of IL-1β, TGF-β1 expression was increased, the result was statistically significant (P<0.05).④tissue detected by superoxide dismutase (SOD) and malondialdehyde (MDA) expression, compared with the model, continuous cold therapy group and intermittent cold therapy group was decreased MDA, SOD expression was increased, the result has statistically significant (P<0.05), compared with intermittent cold therapy, continuous cold therapy group and the group continued intermittent cold therapy cold therapy group was decreased MDA, SOD expression was increased, the result was statistically significant (P<0.05).Conclusion:①Rat model of acute swelling of soft tissue injury gore color change may be related to aseptic inflammation, tissue repair and secondary fiber injury.②Cold therapy can significantly reduce acute soft tissue injury, and cold therapy is superior to intermittent continuous cold therapy.③Cold therapy to reduce acute soft tissue injury may be related to decreased IL-1β, increased TGF-β1, to reduce MDA and increased SOD expression, thereby reducing aseptic inflammation and promote tissue repair and reduce secondary fiber injury.
