Gout is a type of inflammatory arthritis that is triggered by the crystallization of uric acid within the joints and is often associated with Hyperuricemia (HUA). This subject was to screen anti-gout effects and preparation procedure of the compound Chinese medicine of Alismatis Rhizoma and LC and preliminary investigate their possible mechanism action of anti-gout.Acute Hyperuricemia models were successfully cloned by subcutaneous injection of oxonic acid potassium salt and administration of hypoxanthine in mice. Hypouricuria effects of the extracts of Plantaginis Herba were investigated in these models of Hyperuricemia. Orthogonal test design was utilized to optimize the extraction conditions when plantamajoside was selected as the evaluation index. Acute hyperuricemia models were successfully established by administration of uric acid in quails. The therapeutic effects of LC were assessed on rats with acute gouty arthritis (AGA) caused by uric acid crystals. The expression of IL-10 gene was detected by RT-PCR for the further study on anti-inflammatory mechanism of LC. Dose-hypouricuria effect and time-hypouricuria effect of Alismatis Rhizoma and LC were studied on the acute Hyperuricemia models cloned by subcutaneous injection of oxonic acid potassium salt and administration of hypoxanthine in rats. Finally, the preparation process was initially identified by designing and examining the prescription and formulation of the compound traditional Chinese medicine with taking the physicochemical properties of Alismatis Rhizoma and LC into account.The results indicated that the extracts of Plantaginis Herba could significantly decrease the serum uric acid levels of the hyperuricemic mice (P<0.01). All of the concentration of ethanol, extraction duration per time, extraction times and amount of ethanol had significant effects on the content of plantamajoside extracted from Plantaginis Herba. The optimum extraction process was to extract two times using tenfold 60% ethanol and 2 h per time. The degree of the acute gouty arthritis joint swelling in rats could significantly be reduced by LC (P<0.01), IL-10 levels in the articulars significantly increased (P<0.01). The expression of IL-10 gene in rats detected by RT-PCR was strengthened significantly (P<0.01).The traditional Chinese compound medicine of Alismatis Rhizoma and LC could significantly reduce the serum uric acid levels of the hyperuricemic rats (P<0.01) and quails (P<0.01). Lastly, capsules preparation procedure of Alismatis Rhizoma and LC was initially obtained:the recipe were the mixture of Alismatis Rhizoma and LC 100 g, microcrystalline cellulose 10 g, silica powder 4 g; after all of the raw materials were dried respectively, sieved by 80 mesh, wetted by 80% ethanol, granulated with 50 mesh, dried again. The powder of the particles were sieved out by 80 mesh, mixed evenly and filled into the capsules finally, with the CHR controlled below 68%.It could be initially inferred from above-mentioned results that alcohol extract of Plantaginis Herba has hypouricuria effect and plantamajoside might be one of the active ingredients. LC has significant anti-gout effect on acute gouty arthritis and increasing the expression IL-10 gene might be the mechanism. Hypouricuria effect of Alismatis Rhizoma and LC could be validated.
