Objective:This study is to investigate the association of Toll-like receptor 2 gene rs4696480,rs3804099,rs3804100,rs5743704,rs121917864 SNPs with Gouty arthritis(GA);and Hyperuricemia(HUA).Method: Peripheral blood samples from 317 patients with primary gout,385 HUA and 507 healthy subjects were selected as subjects.The TLR2 gene rs4696480,rs3804099,rs3804100,rs5743704,rs121917864 were genotyped by the SNPscanTM multiplex SNP typing kit,analysising the experimental results by Statistical method.Comparison the allele and genotype frequencies in GA,HUA,and healthy controls(HC).Result: 1.The TLR2 gene rs3804099,rs3804100,rs4696480,rs5743704,rs121917864,rs5743708,rs11935252,rs369028698 sites all meet the Hardy-Weinberg equilibrium in the GA,HUA,and HC.2.There were no significant differences in the distribution of 2TLR2 gene rs3804099,rs3804100,rs5743704,rs121917864,rs5743708,rs11935252,rs369028698 locus genotypes and allele frequencies between GA group and HC and between HUA group and HC group(p>0.05).3.There was no significant difference in the distribution of TLR2 gene rs4696480 genotype and allele frequency between GA group and HC group(p>0.05),but the difference between HUA group and HC group was statistically significant.(p<0.05)4.The TLR2 gene rs3804100 and rs3804099 loci were in linkage disequilibrium(r2>0.3,D'>0.8).There was no significant difference in the distribution of CC,CT and TT haplotype frequencies between GA group and HC group(p>0.05).There was no significant difference in the distribution between the HUA group and the HC group(p>0.05).The TLR2 gene rs3804100,rs121917864,and rs5743708 loci were in linkage disequilibrium(r2>0.3,D'>0.8).There was no significant difference in the distribution of CCG and TCG haplotype frequencies between GA group and HC group(p>0.05).There was no significant difference in the distribution between the HUA group and the HC group(p>0.05).Conclusion:1.TLR2 rs4696480 gene polymorphism may be related to HUA,but not to GA.The T allele may be a protective factor for HUA,and carrying the T allele may reduce the prevalence of HUA.2.The polymorphisms of TLR2 rs3804099,rs3804100,rs5743704,rs121917864,rs5743708,rs11935252,and rs369028698 genes may be independent of GA and HUA susceptibility.3.TLR2 gene rs3804099,rs3804100 site CC,CT,TT haplotype and rs3804100,rs121917864,rs5743708 site CCG,TCG haplotype may be unrelated to GAt and HUA.
