Objective: Established IgA nephropathy rat model ,to investigate the effects and the mechanism of Rhizoma Imperatae (RI) and its complex soup on the model of IgA nephropathy in rats. Methods: 50 male SD rats were randomly divided into normal control group, model group, RI group, RI complex group, dexamethasone group and 10 in each . In addition to the normal control group, the other 4 groups were established IgA nephropathy rat model with bovine serum albumin(BSA) plus staphylococcal enterotoxin B (SEB) and carbon tetrachloride (CCl4) subcutaneous injection methods. The continuous administration time was 4 weeks.The normal control group and model group were given normal saline orally.In 8 , 10 and 12 weeks of experiment , the fresh urine of rats were collected to count urinary red blood cell (URBC) and 24 hour urine protein (UP).At the end of 12 weeks, rats were killed to test serum creatinine (Scr), blood urea nitrogen (BUN); The serum interleukin-2 (IL-2) , platelet activating factor (PAF)were tested by enzylne linked immunosorbent assay ( Elisa ) ; Renal specimens from each group were checked by pathological(HE;PASM-Massonstaining;immunofluorescence)analysis; Renal tissue contents of transforming growth factor(TGF-β1) and protein expression were tested by the methods of immunohistochemistry and western blot analysis Results: Compared with the normal control group: at the 8th week, rats of model group began to appear in the microscopic hematuria, microalbuminuria (P<0.01), 10th weeks of significant gross hematuria, proteinuria (P<0.01) ; Scr , BUN levels were significantly increased; serum IL-2 levels were significantly reduced, PAF and TGF-β1 protein expression was significantly increased (P all<0.01); Compared with the model group , the three treatment groups:URBC, 24hUP, Scr, BUN, PAF and TGF-β1 expression were significantly lower, serum IL-2 were significantly increased (P<0.01) ; The dexamethasone group and RI complex group reduced the levels of serum PAF and Scr more obvious than the RI group (P <0.05) ; The RI group and RI complex group elevated levels of IL-2 more obvious than the dexamethasone group (P<0.05). Pathological changes: in the optical microscope: normal control rats were normal glomeruli, the mesangial cells and mesangial matrix of model group were severe hyperplasia, compared with the model group, glomerular mesangial cells and mesangial matrix hyperplasia of the three treatment groups were significantly relieved; In the immunofluorescence: normal control group found no glomerular deposition of IgA fluorescence, model group was a large bolus IgA in glomerular mesangial deposition, the IgA fluorescence intensity of the three treatment groups were lower than model group (P<0.05). Conclusion: RI and its complex soup had a significant therapeutic effect on model of IgA nephropathy, and with the priority of Rhizoma Imperatae complex soup,can significantly reduce hematuria, proteinuria of the IgA nephropathy rats, alleviate the pathological changes, to improve renal function. The mechanism may be the secretion of IL-2 of RI stimulating the body, reduction the production of PAF, inhibition of renal TGF-β1 secretion, expression, etc.
