Objective:To investigate whether Dioscoreae Rhizoma-Imperatae Rhizoma Drug Pair can reduce kidney damage of contrast medium in rats,prevent acute kidney damage caused by contrast medium through experimental research,and try to explore its related mechanism.Methods: Forty-eight healthy clean-grade male SD rats,randomly divided into normal group,model group,Dioscoreae Rhizoma-Imperatae Rhizoma Drug Pair group(Drug Pair group),Dioscoreae Rhizoma group,Imperatae Rhizoma group,acetylcysteine group(NAC group),each grounp had eight rats.Drug Pair group,Dioscoreae Rhizoma group,Imperatae Rhizoma group,NAC group were given daily intragastric administration with corresponding drugs.while normal group and model group were given daily intragastric administration with distilled water for 7 days.Before modeling,each group was banned from water for 12 hours.Except for the normal group,the other groups were replicated with contrast-induced nephropathy.24 hours after the modeling completed,the rats were killed,Renal function was measured,renal structure was observed by HE staining,Caspase-3 and Bcl-2 protein expression were detected by immunohistochemistry,and renal tubular epithelial cells apoptosis was detected by TUNEL,the apoptosis index(AI)aslo be calculated.Result : 1.Biochemical indexes:compared with normal group,the level of Scr ? BUN in the model group were significantly increased(p <0.05).Compared with model group,the level of Scr?BUN in the Drug Pair group and NAC group were decreased significantly(p < 0.05),while the level of Scr ? BUN in the Dioscoreae Rhizoma group Imperatae Rhizoma group no significant decreased;2.Pathological observation:The renal tubular morphology of the rats in the normal group was basically normal.In the model group,the renal tubules were significantly dilated,the tubular wall was damaged,the structure was unclear,vacuole cells were formed,and red blood cell casts were formed.Renal lesions were alleviated especially in the Drug Pair group and NAC group,while in the other groups the structure has no obvious changes.3.Immunohistochemistry:Bcl-2:Compared with the normal group,Bcl-2 protein in the model group especially decreased(p < 0.05).Compared with the model group,the expression of Bcl-2 in Drug Pair group and NAC group were significantly increased(p < 0.05),there was no difference between the two groups,and there was no significant difference between tthe model group and the other treatment groups(p > 0.05);Caspase-3:Compared with the normal group,the expression of Caspase-3 protein in the model group was significantly increased(p< 0.05),Compared with the model group,Caspase-3 protein in Drug Pair group and NAC group were significantly reduced(p< 0.05)and there was no significant difference between model group and other treatment groups(p > 0.05).4.Apoptosis index(AI):Compared with the normal group,the AI in the model group was significantly increased(p<0.05),the AI in Drug Pair group and NAC group was significantly reduced(p <0.05),AI in other treatment groups was reduced but not statistically significant(p>0.05).Conclusion: Dioscoreae Rhizoma-Imperatae Rhizoma Drug Pair can improve the renal function and kidney structure damage of contrast-induced nephropathy model rats,up-regulate the expression of Bcl-2 protein,down-regulate the expression of Caspase-3 protein,reduce renal tubular epithelial cell apoptosis and finally protect the kidneys and prevent the kidney damage caused by contrast medium.
