| Imperatae Rhizoma is a commonly used Chinese herb.It has function of cooling blood and hemostasis,clearing heat and diuresis,and is commonly used to treat acute nephritis,acute infectious hepatitis.Polygonati odorati Rhizoma is also a Chinese herb.It has the effect of nourishing yin to moisten dryness and helping produce saliva to slake thirst.It is commonly used to treat lung and stomach yin injury,cough caused by dryness-heat,dry throat and thirst,internal heat dispersion-thirst.Their quality standards were recorded in Chinese Pharmacopoeia(the version of 2015).But there is a lack of exclusive and characteristic content determination method,which is not specialized to effective quality control.Based on the current Chinese Pharmacopoeia standards,samples collected by resource census were studied to improve the control level of quality standards.To achieve the quantitative analysis of the organic acids in Imperatae Rhizoma,a HPLC method was established for determination of chlorogenic acid and a HPLC fingerprint method was also developed meanwhile,which the similarity value was calculated by comparison with that of reference crude herb.The results showed that the content of chlorogenic acid in the 56 samples were in the range of 0.018~1.497mg.g-1 and the similarity value of samples were different.Samples with low similarity had also low chlorogenic acid content.Under the condition that the similarity value is less than 0.5 and the content of chlorogenic acid is less than 0.05mg.g-1,most of the suspected samples can be excluded.To clarify the existence of the main carbohydrate in Rhizoma Imperatae,a HPLC-CAD method was used to determine the content of fructose,glucose and sucrose,and clustering analysis of the content was also carried out.The results showed that there was a correlation between the content of these three carbohydrates and sample quality,and it can be used to evaluate the quality of Imperatae Rhizoma and to distinguish the unqualified samples.According to results,three kinds of carbohydrate in the qualified Imperatae Rhizoma were determined,the content of fructose should not be less than 2%,the content of glucose not less than 2%,and the content of sucrose should not be less than 5.5%.To figure out the characteristics of polysaccharide in Imperatae Rhizoma,the HPGPC method was used to determine the molecular weight and distribution of polysaccharides.The results showed that among the 2 main chromatographic peaks in HPGPC graphs,peak 1 was macromolecule polysaccharide,peak2 was oligosaccharide with smaller molecular weight,and the peak area of peak2 was much larger than that of peakl.This result indicated that the content of oligosaccharide was higher than that of polysaccharide with high molecular.The HGPC-RI-MALLS method was used to determine the polysaccharide of Imperatae Rhizoma.The molecular weight of polysaccharide was determined directly without the standard substance.At the same time,the molecular rotation radius of polysaccharide was obtained,the molecular characteristics of the polysaccharide in Rhizoma Imperatae were further revealed.The components of carbohydrates in Polygonati odorati Rhizoma were analyzed and determined by HPLC-CAD method.The results showed that fructose and sucrose were present in most of the samples,and glucose was not detected in individual samples.The content of fructose in Polygonati odorati Rhizoma was in the range of 9.585~88.561mg.g-1 and the content of sucrose was lower in the samples with smaller diameter.The molecular weight and distribution of polysaccharides in Polygonati odorati Rhizoma was determined by HPGPC method and HGPC-RI-MALLS method.The indexes suitable for characterization of polysaccharides were obtained.There were three main chromatographic peaks in HPGPC graphs,peakl was the large molecular weight polysaccharide,peak2 was the small molecular weight polysaccharide,and peak3 was the small molecule oligosaccharide.The results showed that the main chromatographic peaks had certain characteristics,and the peak area of peak 2 should be larger than that of peak 3,meanwhile the Mw range of peakl was 50000 to 125000Da,the Mw of peak2 was 2500 to 5000Da,and the distribution index was less than 1.6.The analysis result of HGPC-RI-MALLS method provided the information of molecular weight,molecular rotation radius and other molecular characteristics of polysaccharide,which can be used as reference for the future research of polysaccharide in Polygonati odorati Rhizoma.Based on the above research results,new standard monograph drafts of the two herbs were written separately,including new items of quality control. |
