Objective:1 To establish the best water extraction and alcohol precipitation processes for MaTengDingChuanFang.2 To select Pharmacological active site preliminary of MaTengDingChuanFang for antitussive and preventing asthma.3 To set up the content determination method of ephedrine hydrochloride in Effective components group by HPLC.4 To study the acute toxic reaction of Effective components group from MaTengDingChuanFan in mice.Methods:1 Orthogonal test applied to extraction procedure for MaTengDingChuanFang. Choose the content of Ephedrine Hydro-chloride as an index which was determined by HPLC.2 The cough model guided by thick ammonia waz employed to determine effective parts of antitussive, the asthma model guided by phosphoric acid histamine and acetylcholine mixture waz employed to determine effective parts of relieving asthma.3 HPLC method was used with Diamonsil C18 column (250 mm X4. 6 mm,5μm). The flowing phase was 0.1% phosphoric solution: acetonitrile (94:6) with f low rate of 1.0 mL·min-1. T he detecteing wavelength was at 220nm,, the column temperature was thirty.4 We fed the mice with the maximum administration concentration drug for 7 days, and gavage administration was done for three times within 24 h. Then, we observed the action, state, diet, hair color, stool, secretion and death amount of the mice.Results:1 The optimal water extraction process was added 10 times water for 3 times and 1.5 hours for each time. The best alcohol precipitation techn ique was as follows:60% of precipitation concentration, the alcohol deposit continued 24 h, the enrichment ratio was 1:1.5.2 The 95% and70% water/ethanol parts from MaTengDingChuanFang have got obviously relieving cough function against mouse cough guided by thick ammonia;and had evident inhibition on guinea asthma induced by phosphoric acid histamine and acetylcholine mixture.3 Within the range of 0.32~0.74μg, the linear relationship was good for ephedrine hydrochloride (r=0.9999),the rate of average recovery was 96.4%, RSD= 1.92%(n= 6).4 All mice survived and no toxic reaction was shown in the observed period. The maximum administration dosage of Effective components groupfrom MaTengDingChuanFan was 19.68g/kg, converting into the crude drug was 1100.112g/kg, equivalent to men's daily administration dosage by 612 times.Conclusion:1 The optimized extraction is stable, economical and feasible, and the HPLC method for its determination is simple, rapid and accurate.2 The 95% and70% water/ethanol elution parts from MaTengDingChuanFang is the effective portion of antitussive and ant iasthmatic.3 The method was accurate, simple and practicable with good reproducibility for determination of Ephedrine Hydrochloride in Effective components group from MaTengDingChuanFang.4 The effective components group from MaTengDingChuanFan is secure for oral administration, it was safe for clinical use.
