Objectives:To investigate the expression of Nanog and Oct-4 gene in BPH,PIN and PCa, analysis its effect on the development of the disease and its clinical significance.Methods:In this study, expression of Nanog and Oct-4 was detected by immunohistochemistry analysis in 132 patients,30 had BPH,8 had PIN,94 had prostate cancer (PCa). And between these PCa patients, there are 23 of them GleasonScore below 4',42 of them GS between 5'to 7',and 29 of them GS higher than 8'. Associations with various clinical pathological prognostic variables were analyzed in 94 PCa patients.Results:1. According to PC-3 and LNCaP Immunofluorescence test, there are obvious red fluorescence in both PC-3 and LNCaP cell nucleus, we also observe weak red fluorescence in a few prostatic cell cytoplasm.2. Expression levels of Nanog and Oct-4 were higher in PCa (100% and 95.7%) tissues than in PIN (25% and 50%) tissues; Expression levels of Nanog and Oct-4 in PIN tissues were slightly stronger than BPH (16.7% and 10.0%) tissues.3. There are positive relationship between Gleason Score and the expression of Nanog or Oct-4. Associations with various clinical pathological prognostic variables such as patients'age and TPSA seems no obvious relationship with the expression of Nanog or Oct-4.Conclusions:Nanog and Oct-4 expression levels showed significantly differences according to different PCa stage. Our findings indicate that Nanog and Oct-4 may be involved in carcinogenesis of the prostate and progression of PCa.
