BackgroundThere are a lot of different kinds of stem cells in myocardial tissue,which are of great significance for the regeneration and repair of myocardium.The pericardium is very similarity to the myocardium in the occurrence,and as a peripheral organ of the heart,there should be similar cell composition in the theory.However,it is not clear whether there are subsets of cardiac stem cells.ObjectiveTo study the where is the distribution of c-kit,Sca-1 and Nanog positive cells in the pericardium,and what is the relationship between them.To help people use stem cell therapy for cure acute myocardial infarction.Methods1.The rats were sacrificed by cervical dislocation,then take off their fresh pericardium.Frozen section and paraffin section technology were used to make slices.HE and Masson were used to stain.2.Immunohistochemical staining was used to identify the presence and distribution of c-kit positive cells,Sca-1 positive cells and Nanog positive cells in the pericardium.3.The expression of c-kit positive cells,Sca-1 positive cells and Nanog positive cells in the pericardium and the relationship between them were demonstrated by immunofluore-scence staining.4.We use western blot technology to observe the expression of c-kit,Sca-1 and Nanog protein in pericardium,and to make a semi quantitative analysis of them.Results1.Histologically,the pericardium is composed mainly of fibrous connective tissue,which contains small,round cells.Immunohistochemistry showed that c-kit positive cells,Sca-1 positive cells and Nanog positive cells were found in the pericardium.2.The c-kit positive cells and Sca-1 positive cells can express the same surface markers,Sca-1 positive cells and Nanog cells can express the same surface markers,c-kit positive cells and Nanog positive cells can express the same surface markers.3.The relative expression of c-kit protein,Nanog protein and Sca-1 protein was 0.88± 0.08,0.68 ± 0.05,0.87 ± 0.09.ConclusionsThere were three kind of positive cell subsets of c-kit,Sca-1 and Nanog in the pericardium,and there is cross expression between them.These cells are the same as those of cardiac stem cells,maybe it will play an important role in myocardial regeneration.
